Do NAAT-based methods increase the diagnostic sensitivity of streptococcus agalactiae carriage detection in pregnant women?

The aim of the study was to evaluate particular polymerase chain reaction primers targeting selected representative genes and the influence of a preincubation step in a selective broth on the sensitivity of group B Streptococcus (GBS) detection by nucleic acid amplification techniques (NAAT). Research samples were vaginal and rectal swabs collected in duplicate from 97 pregnant women. They were used for enrichment broth culture-based diagnostics, bacterial DNA isolation, and amplification, using primers based on species-specific 16S rRNA, atr and cfb genes. To assess the sensitivity of GBS detection, additional isolation of samples preincubated in Todd-Hewitt broth with colistin and nalidixic acid was performed and then subjected to amplification again. The introduction of the preincubation step increased the sensitivity of GBS detection by about 33–63%. Moreover, NAAT made it possible to identify GBS DNA in an additional six samples that were negative in culture. The highest number of true positive results compared to the culture was obtained with the atr gene primers, as compared to cfb and 16S rRNA primers. Isolation of bacterial DNA after preincubation in enrichment broth significantly increases the sensitivity of NAAT-based methods applied for the detection of GBS from vaginal and rectal swabs. In the case of the cfb gene, the use of an additional gene to ensure the appropriate results should be considered.

According to many surveys of pathogenic organisms in forest soils, the presence of the Phytophthora genus is very common in both dominant and mixed stands of European beech. In Serbia, Phytophthora species were isolated from rhizosphere soil in declining, as well as apparently healthy stands. After detailed morphological and molecular identification, several Phytophthora species were confirmed. The most common pathogen of fine roots in Serbian European beech stands was Phytophthora plurivora Jung and Burgess. This species was characterized as homothallic, semipapillate, produces sporangia of various shapes, and has an optimum temperature for growth at around 25ºC. P. plurivora occurred on 58% of positive samples, followed by P. cambivora (Petri) Buisman at 17%, P. gonapodyides (Petersen) Buisman at 8%, with other unidentified species accounting for the remaining 17%. A pathogenicity test performed with P. plurivora and young beech germinants from ten Polish beech provenances demonstrated the ability of this pathogen to colonize and cause deterioration of plant tissue.

Cloacal and pharyngeal swabs were sampled from 33 Alpine Accentors. A total of 32 specimens were Yersinia positive, with 73% of birds being positive for Y. enterocolitica and 51% for Yersinia spp. A comparison of host characters and environmental conditions showed these to be consistent with the different life strategies of Y. enterocolitica and other Yersinia species. Y. enterocolitica is more successful at colonizing the birds' digestive tracts — the occurrence of Y. enterocolitica was significantly higher in the cloacal than the pharyngeal swabs. The occurrence of Y. enterocolitica was high in summer, especially in the nesting period (July). In juveniles (including nestlings), there was a 100% prevalence of Y. enterocolitica, whereas only two out of nine juveniles were Yersinia spp. positive. There was no significant difference between the occurrence of Y. enterocolitica in anthropogenic and natural habitats, but the occurrence of Yersinia spp. was much greater in the former than in the latter habitats. The presence of the ail gene associated with pathogenic Y. enterocolitica strains was not confirmed in any of the samples examined.
