Disruption of the neuronal membrane by toxic amyloid β oligomers is hypothesized to be the major event associated with Alzheimer's disease's neurotoxicity. Misfolding of amyloid β is followed by aggregation via different pathways in which structurally different amyloid β oligomers can be formed. The respective toxic actions of these structurally diverse oligomers can vary significantly. Linking a particular toxic action to a structurally unique kind of amyloid β oligomers and resolving their toxicity-determining feature remains challenging because of their transient stability and heterogeneity. Moreover, the lipids that make up the membrane affect amyloid β oligomers' behavior, thus adding to the problem's complexity. The present review compares and analyzes the latest results to improve understanding of amyloid β oligomers' interaction with lipid bilayers.

In the gap of stability : the search for old media business models in the digital environment

For traditional media companies such as radio, television and especially the Press, the formation of effective business models in the Internet ensuring stable functioning in digital environment is a fundamental strategic challenge. Over the years, these companies followed their readers and viewers into the so-called new media environment. However, their engagement in the Internet activity outpaced understanding of how to benefit from it financially, at least to compensate its costs. From a strategic point of view, digital distribution channels remain a risky, though necessary, strategic decision for traditional media companies. Media companies, especially publishers, which use old models based primarily on advertising revenues have been standing in the transitional period of “stability gap” for more than 20 years due to the lack of continuity of effective business solutions. They are trapped in this transitional period when the existing models no longer provide development, and new effective solutions either do not exist yet, or are exceptions unable to be adapted creatively. In face of common threats of revenues decline, efforts to seek fresh models focus on the idea of new aggregation. Few successful examples in various global markets, however, require creative development in the diverse media systems.

Host-derived protease promotes aggregation of Staphylococcus aureus by cleaving the surface protein SasG

Staphylococcus aureus is one of the leading causes of hospital-acquired infections, many of which begin following attachment and accumulation on indwelling medical devices or diseased tissue. These infections are often linked to the establishment of biofilms, but another often overlooked key characteristic allowing S. aureus to establish persistent infection is the formation of planktonic aggregates. Such aggregates are physiologically similar to biofilms and protect pathogens from innate immune clearance and increase antibiotic tolerance. The cell-wall-associated protein SasG has been implicated in biofilm formation via mechanisms of intercellular aggregation but the mechanism in the context of disease is largely unknown. We have previously shown that the expression of cell-wall-anchored proteins involved in biofilm formation is controlled by the ArlRS-MgrA regulatory cascade. In this work, we demonstrate that the ArlRS two-component system controls aggregation, by repressing the expression of sasG by activation of the global regulator MgrA. We also demonstrate that SasG must be proteolytically processed by a non-staphylococcal protease to induce aggregation and that strains expressing functional full-length sasG aggregate significantly upon proteolysis by a mucosal-derived host protease found in human saliva. We used fractionation and N-terminal sequencing to demonstrate that human trypsin within saliva cleaves within the A domain of SasG to expose the B domain and induce aggregation. Finally, we demonstrated that SasG is involved in virulence during mouse lung infection. Together, our data point to SasG, its processing by host proteases, and SasG-driven aggregation as important elements of S. aureus adaptation to the host environment.

Despite the fundamental importance and broad applicability of E/Z dicarboxylic acids, their discrimination remains challenging and greatly unexplored. Herein, we present a general approach for the recognition of E/Z diacids using supramolecular interactions coupled with plasmonic response. The method allows detecting both single isomers and their light-induced interconversion, which ultimately entails multiple reversible nanoparticle aggregations. Such a molecular recognition-coupled responsive nanoscale self-assembly resembles natural mechanisms and can be a versatile means of building artificial complexity.

Convergence has contributed to placing some media contents on different electronic platforms. Bloggers and internet users have almost taken professional journalist’s place. The evolution has affected all branches of media sector, the citizen journalism has entered into the new media and social networks have become a source of information for many people. New models of media content management are primarily targeted at reducing costs and increasing profits and at splitting off from the traditional bilateral management model. In the modern chaos of information overload society needs to have not only free, clear and segregated media content available from a variety of devices but also to share their opinion with others. Such demand was sensed by Arianna Huffington who created the whole media empire counting more than 100 million readers from all over the world. She has created a huge opinion-forming platform (The Huffington Post) which combines a news aggregator and blogging platform together to which you can add TV channels, magazines and e-books activity. Such combination of media content aggregator activity with bloggers platform activity can be defined as “huffinization”. Media content are created by the platform users and more than 30 000 unpaid bloggers that is allowed not only to ramp up profits but also stimulate the development of political and social events.
