TLC determination of meloxicam in tablets and after acidic and alkaline hydrolysis

Opis:
A simple and rapid method for separation and determination of meloxicam and its degradation products by thin-layer chromatography with densitometric detection in pharmaceutical preparations was described. The method employed TLC F254 plates as the stationary phase. The solvent system consisted of ethyl acetate : toluene : butylamine (2:2:1, v/v/v). Densitometric analysis was carried out in absorbance mode at wavelength of 297 nm. The method was validated for linearity, precision and accuracy. The limits of detection and determination were 0.96 μg per spot and 2.90 μg per spot, respectively. The drug was degraded in acidic and basic environment, at different temperatures. The degradation products were well resolved from the active substance. The HPLC-MS/MS method for the identification of degradation products of meloxicam (i.e. 5 methylthiazol- 2-ylamine and 5-(dioxide-l6-sulfanylidene)-6-methylidenecyclohexa-1,3-diene) was investigated. Because the presented method allows the efficient separation of the drug from some of its degradation products, so it can be used as a stability-indicating analysis.

The article presents the validation procedure of the questionnaire to measure individual readiness to resist against unwanted social impact. One of the aims of the research was to design a tool for use in the academic education context. The conceptualization was based on literature relating to rebelliousness, negativism, reactance, disobedience and resistance against social influence. Consequently, it has been assumed that the variable is multidimensional. Results show that the tool measures four separate types of readiness to resist. Moreover, the validity and the reliability were satisfactory. The empirical data was collected in a series of investigations on a group of 653 people.

Psychometric validation of the polish version of the emotional style questionnaire

In three studies, we document the validity of the Polish version of the Emotional Style Questionnaire (ESQ)—a 24-item self-report measure that captures how people vary across six dimensions that make up a healthy emotional life. Study 1 provides support for the proposed factorial structure of the Polish version of the scale and gender measurement invariance, demonstrates the similarity of the Polish and English versions of the scale, and reveals the excellent test-retest reliability of the scale across an interval of two months. Study 2 confirms the adequacy of the factorial structure in another sample, shows that the construct validity for each of the six subscales is similar to that of the original scale, and documents the age measurement invariance of the scale. Finally, in Study 3, we investigate measurement invariance across Polish and American samples and demonstrate that Poles are characterized by lower scores on healthy emotionality and its dimensions compared to Americans. We conclude that the Polish version of the ESQ is a psychometrically valid and easily implementable measurement tool that can be applied in research settings to measure healthy emotionality and its components.

Determination of linezolid in human serum by reversed-phase high performance liquid chromatography with ultrafiolet and diode array detection

A high-performance liquid chromatographic (HPLC) method with UV and DAD detection for the quantitative determination of linezolid in human serum was developed in present work. Chromatography was carried out by reversed-phase technique on a RP-18 column with a mobile phase composed of 50 mM phosphate buffer and acetonitrile (76 : 26, v/v), adjusted to pH 3.5 with orthophosphoric acid. Serum samples were deproteinized with methanol centrifuged and then, the supernatant was analyzed using HPLC procedure. No interference was observed at the retention times of linezolid from blank serum or ten commonly used antibiotics. A concentration range from 0.50 to 30.0 g/mL was utilized to construct calibration curves. The lower limit of detection was determined to be 0.1 μg/mL of serum for both detectors. The lower limit of quantification of 0.25 μg/mL (CV = 2.6%) was established for determination using HPLC-UV and 0.5 μg/mL (CV = 5.42%) for HPLC-DAD. The recovery of linezolid was approximately 100%. Intra-day accuracy ranged from 0.97 to 12.63% and 0.74 to 10.85% for HPLC-UV and HPLC-DAD method, respectively. Intra-day precision was less than 4.69% for HPLC-UV and less than 5.42% for HPLC-DAD method. Tests confirmed the stability of linezolid in serum during three freeze-thaw cycles and during long-term storage of frozen serum for up to 6 weeks; in extracts it was stable in the HPLC autosampler over 24 h. Statistical analysis by Student's t-test showed no significant difference between the results obtained by these two methods. In summary, these methods will be used and adapted for infected patients in intensive care unit, to determine linezolid serum concentrations in order to know the pharmacokinetic profiles of linezolid.

Fear of falling is associated with a clear hazard to individual self-reliance, reduced physical activity, as well as a sense of shame and loss of self-confidence. The present study aimed to complete the applicable translation and validation protocol for the Falls Efficacy Scale—International (FES-I) tool, following its prior adaptation to ensure full compatibility with the Polish setting. The FES-I questionnaire, along with its abridged version, was translated in line with the recommended standards of the MAPI Institute, taking into account both the cultural fabric and pertinent language specifics of the country. The survey was attended by 740 individuals (N = 740; 463 women, 277 men), over 60 years old. All respondents were required to complete both the FES-I and FES-I (Short) questionnaires twice, following an intervening period, and subsequently had their responses statistically assessed. The FES-I questionnaire, along with its abridged version, may be recommended as an effective assessment tool for addressing the fear of falling issue among the older adults, consequently allowing the teams of attending physicians, physiotherapists, psychologists, or psychiatrists to complete an unambiguous diagnosis, with a view to helping the patients overcome this particular type of anxiety.
