Opis:
Blastocystis spp. is a parasite that causes intestinal infection in humans and other animals. A few studies have been performed in Turkey on the distribution of Blastocystis in cattle. In this study, fecal samples were collected from 100 calves and subjected to analysis based on an SSU rRNA gene fragment. The overall prevalence of the disease was determined as 15% (15/100). This rate was 14.04% for females and 16.28% for males. In addition, three Blastocystis subtypes were identified: ST10, ST14, and novel subtypes ST25. To our knowledge, the ST25 subtype was reported with this study for the first time in Turkey. The nucleotide sequences (OM920832-OM920839) obtained in this study were deposited in GenBank. The results obtained will be useful for a better understanding of the epidemiology of Blastocystis spp., and its effects on public health.

Opis:
Globally, more than 5.2 billion hectares of farming fields are damaged through erosion, salinity, and soil deterioration. Salt stress-tolerant bacteria have plant growth-promoting (PGP) characteristics that can be used to overcome environmental stresses. Isolation and screening of salt-tolerant endophytes from Bougainvillea glabra were achieved through surface sterilization of leaves, followed by their cultivation on 0.5% NaCl-supplemented media. The performance of isolates for indole acetic acid (IAA) production, phosphate solubilization, ACC deaminase activity, ammonia production, siderophore production, and stress tolerance was determined. In the present study, 5 morphologically distinct salt-tolerant endophytic bacteria were cultured. Out of 5 isolates, 60% salt-tolerant endophytes showed phosphate solubilization, 100% IAA production, 40% ACC-deaminase activity, 100% siderophore production, 40% ammonia production and 60% HCN production. Dendrogram generated based on stress tolerance showed three clusters: clusters 1 and 2 containing two isolates and cluster 3 containing 1 isolate. Based on the highest PGP activities, BoGl17 and BoGl21 isolates were chosen and further tested for wheat and corn growth promotion. The isolates increased wheat shoot and root dry weight by 40% and 80%, and by 69% and 126% for BoGl17 and BoGl21, respectively. The bacterial isolates identified as Pseudomonas reidholzensis BoGl17 had 95.59% identity with Pseudomonas reidholzensis strain ID3, while the isolate Bacillus aerius BoGl21 had 90.96% identity with Bacillus aerius strain 24K based on phenotypic features and 16S rRNA gene sequencing results. Both isolates significantly improved plant growth as compared to uninoculated plants.

Shrimp activity is associated with the impact of bacterial communities. Therefore, this research aimed to evaluate the influence of the bacterial microbiota on the organic matter content of the soil of the shrimp lagoon in La Segua-Ecuador. Starting from a descriptive approach, the field research method and documentary review were used. In total, 25 soil samples were collected in 5 quadrants of 100 m2. The bacterial DNA was extracted by using the Powersoil® kit and the identification of the strains was carried out with the 16SrDNA gene. The organic matter content was determined by Walkley-Black titration. The genus Bacillus was predominant in the bacterial strains; moreover, individuals of the genera Exiguobacterium, Acinetobacter, Prolinoborus, Arthrobacter Planococcus were identified with more than 99% homology for all cases. It was concluded that the organic matter content is suitable for shrimp farming.

The molecular analysis of 32 durian F1 hybrids, resulted from crossing of the Arp 8990 (female parent) and ‘Otong’ (male parent), was conducted in order to determine the genetic characteristics of hybrids and parents, as it would be followed/evidenced by the variability of traits produced from the cross breeding. The RAPD analyses of 14 primers resulted in 114 scoring bands, 112 (98.2%) of them were polymorphic, with 4 to 11 bands amplified per primer. The electrophoresis gel of the PCR results revealed that some hybrids produced different band patterns compared to the parents; this indicated the crossing between parents’ alleles and trait combinations from both the parents. The Dice-Sorensen similarity coefficient demonstrated that most of the hybrids had distant genetic similarities with both parents, which were ranged from 0.141 [71B(4) and 72B(15)] to 0.776 [71B(15) and 48B(1)]. The UPGMA method was used to construct the dendrogram, which grouped the hybrids in five clusters with distinct genetic relationships and was confirmed with the PCA analysis. This result implied that above crossing produced hybrids having characters different from the parents.

Arnica montana L. is one of the most important herbal plants used in medicine, pharmaceutical and cosmetic industry. The number of studies performed with molecular markers on arnica genotypes is very limited. Because of this fact the aims of presented examination were optimization of protocols DNA isolation from fresh leaves of A. montana and identification of genetic diversity among this plant genotypes. In presented study to obtain pure DNA Plant & Fungi DNA Purification Kit (EURx) were used. To clean obtained DNA long and slow electrophoresis and isolation DNA from gels were used. A. montana genotypes were analyzed using 40 RAPD primers (Operon Technologies), out of which 12 produced high number of polymorphic and repeatable fragments. In total, selected primers produced 120 fragments, among them 111 (92.5%) were polymorphic. The genetic similarity matrices were produced based on RAPD using the Dice’s coefficient. RAPD based genetic similarity was estimated between 0.535 and 0.945. The highest genetic similarity was estimated among GA17 and GA18 genotypes, which are closely located on the obtained dendrogramme.
Arnica montana L. jest jedną z najcenniejszych roślin zielarskich wykorzystywanych w medycynie, farmacji i przemyśle kosmetycznym. W dostępnej literaturze liczba doniesień związanych z analizą molekularną arniki jest znikoma, dlatego też celem prezentowanych badań była optymalizacja procesu izolacji DNA ze świeżych liści oraz identyfikacja zróżnicowania genetycznego oparta na markerach RAPD. W prezentowanej pracy w celu uzyskania czystego DNA do izolacji wykorzystano zestaw DNA Plant & Fungi DNA Purification Kit (Euro) oraz oczyszczanie za pomocą długiej elektroforezy w żelu agarozowym. Spośród testowanych 40 starterów RPAD do analiz wybrano 12 generujących stabilne i polimorficzne wzory prążków. Wyselekcjonowane startery amplifikowały 120 fragmentów, spośród których 111 (92,5%) było polimorficznych. Wykorzystujac markery RAPD utworzono matryce podobieństwa genetycznego. średnia wartość podobieństwa analizowanych genotypów wynosiła 0.886. Najwyższy współczynnik podobieństwa genetycznego oszacowano pomiędzy genotypami GA17 i GA18, które ulokowały się blisko siebie na uzyskanym dendrogramie.

The aim of this study was to perform molecular analysis of canine adenovirus 2 (CAV-2) E1B 19K gene fragment isolated from 20 dogs of various breeds (12 males and 8 females aged 1-9 years), with clinical symptoms of upper respiratory tract infections, from the Lubelszczyzna region. Nasal swabs were taken from dogs. DNA of CAV-2 was detected using the PCR method in 16 swabs. All PCR products were sequenced, and the obtained sequences were compared with each other and with the sequence of the E1B 19K gene of the CAV-2 strain from an online database of NCBI GenBank: AC 000003. Based on analysis of the obtained sequences, three polymorphic variants of CAV-2 (No.1-3) with homology of 78 - 100% were distinguished. The nucleotide and amino acid sequences of the most frequently represented polymorphic variant, No. 1, differed from the sequences of polymorphic variant No. 2 with one substitution. The nucleotide and amino acid sequence of the E1B 19K gene of CAV-2 AC 000003 differed from the analogous sequences of representatives of variant No. 1 with 44 nucleotide and 19 amino acid substitutions. The small number of nucleotide differences in the E1B 19K CAV-2 gene among the examined own isolates, compared with AC 000003, suggest that the infections in dogs were caused by a relatively genetically stable virus which occurs in eastern Poland.
