Opis:
Quantitative description of the interaction between doxorubicin (DOX), a broadly used anticancer drug, and DNA is the key to understand the action mechanism and side effects of its clinical use. However, the reported equilibrium constants of DOX–DNA interaction obtained using a range of different analytical methods vary even by several orders of magnitude. Herein, we propose a novel application of a single-molecule technique – fluorescence correlation spectroscopy (FCS) – to probe the interaction between DOX and two types of DNA (pUC19 and calf thymus DNA), taking advantage of intrinsic self-fluorescence of DOX. We provide an analytical formula for autocorrelation analysis to determine the equilibriumconstant of DOX–DNA complex-formation, where binding of multiple DOX molecules to a DNA chain is included in the reaction–diffusion model. Our FCS-based method not only quantitatively revealed the values of equilibrium constant, but also implied that the stability of DOX–DNA complex is related to the types of base pair rather than the length or structure of the DNA. This work opens a promising pathway toward quantitative determination of molecular interactions in complex systems such as living cells or organisms at single-molecule level.

Opis:
The experimental UV–Vis spectra of the biologically relevant [2Fe–2S] iron–sulfur clusters feature typically three bands in the 300–800 nm range. Based on ground-state orbitals and using the one electron transition picture, these bands are said to be of charge transfer character. The key complication in the electronic structure calculations of these compounds are the antiferromagnetic coupling of the iron centers and high covalency of Fe–S bonds. Thus, the examples of the direct computations of electronically excited states of these systems are rare. Whereas low lying electronic excited states were subject of recent studies, higher energy states computed with many-body theories were never reported. In this work we present, for the first time, calculations of the electronic spectra of [Fe2S2](SMe) 2− 4 biomimetic compound. We demonstrate that spin-averaged restricted open-shell Hartree–Fock orbitals are superior to high-spin orbitals and are convenient reference for subsequent configuration interaction calculations. Moreover, the use of conventional configuration interaction methods enabled us to study the nature of the excited states in details with the difference density maps. By systematic extension of the donor orbital space we show that key excitations in the 300–800 nm range are of Fe 3d ← (μ-S) character.

Rènao: What does it mean to have a good time the Chinese way? An ethnopragmatic exploration of a Chinese cultural keyword

The paper examines the main uses and the symbolic significance of the Chinese cultural keyword rènao. Often rendered in English with its literal gloss of ‘hot and noisy’, it has been viewed by both Chinese and Western scholars as primary in making sense of Chinese social behaviour, across a variety of contexts. The present study analyses two Chinese cultural texts – a report from a local temple festival and a debate over two different styles of feasting, which frequently rely on this salient cultural notion. While the formula crowds, events, noise in the psychological literature dealing with this Chinese social value is often confirmed by the described cultural data, it is argued that close attention to the meaning and form of the descriptive language used by the cultural actors yields valuable insights into indigenous viewpoints. In particular, the notion of rènao turns out to be closely intertwined with other prominent Chinese cultural concerns, such as the idea of rénqíngwèi (‘flavour of human feelings’), Chinese cultural identity, Chinese language, and a particularly complex culinary culture as described in the anthropological literature.

Opis:
In the studies on selection and population genetics of forest trees that include the analysis of genotype × environment interaction (GE), the use of biplot graphs is relatively rare. This article describes the models and analytic methods useful in the biplot graphs, which enable the analyses of mega-environments, selection of the testing environment, as well as the evaluation of genotype stability. The main method presented in the paper is the GGE biplot method (G – genotype effect, GE –genotype × environment interaction effect). At the same time, other methods have also been referred to, such as, SVD (singular value decomposition), PCA (principal component analysis), linear-bilinear SREG model (sites regression), linear-bilinear GREG model (genotypes regression) and AMMI (additive main effects multiplicative interaction). The potential of biplot method is presented based on the data on growth height of 20 European beech genotypes (Fagus sylvatica L.), generated from real data concerning selection trials and carried out in 5 different environments. The combined ANOVA was performed using fixed-effects, as well as mixed-effects models, and significant interaction GE was shown. The GGE biplot graphs were constructed using PCA. The first principal component (GGE1) explained 54%, and the second (GGE2) explained more than 23% of the total variation. The similarity between environments was evaluated by means of the AEC method, which allowed us to determine one mega-environment that comprised of 4 environments. None of the tested environments represented the ideal one for trial on genotype selection. The GGE biplot graphs enabled: (a) the detection of a stable genotype in terms of tree height (high and low), (b) the genotype evaluation by ranking with respect to the height and genotype stability, (c) determination of an ideal genotype, (d) the comparison of genotypes in 2 chosen environments.

In the article we make an analysis of a thesis that verbal symbolic interaction is a necessary condition of constructing self. The main concepts used in the paper are: symbolic interaction, self and corporality. The aforementioned thesis and the concept of symbolic interaction originate from G.H Mead, who set the trend of thinking about interaction in human society in sociology and social psychology. This influence is noticeable up to this day. Symbolic interaction as a tool of understanding others actions and informing partners about our intensions is clearly visible in “languagecentred” and anthropocentrically oriented analyses of interactions as well as in the concentration on linguistic conditions of creating a self. Self is understood as an interpreted concept of a person but mainly in a process of social perception of a human by others occurring in interactions based on verbal language. In the article we want to develop a thesis about “nonlinguistic” possibilities of constructing interactions and self. The aforementioned thesis has been many times elaborated so far together with critical analyses of G. H. Mead (Irvin, 2004, Sanders, 1993, 1999, 2003; Myers, 1999, 2003). We want to integrate these elaborations, including our empirical experiences from a research on “The Social World of Pet’s Owners’ (research done in 2001-2005) on theoretical level and concentrate more on corporality and emotions issues and their relations to symbolic interaction and self. G.H. Mead’s views on this topic are analysed with regard to their methodological consistency and adequacy. In the article there is another thesis proposed, that interactions between animals also have meanings and, sometimes, symbolic nature, or sometimes, non symbolic one, and not necessarily related to use of a verbal language. The creation of self is connected with issues of corporality that includes: 1. nonverbal communication, 2. a relation of bodies in physical space, 3. the so called “kinesthetic empathy”, 4. emotions connected with body, mind and self processes. These elements of corporality may be the basis for taking the role of other. Researches and analyses of many sociologists (beginning from Ch. H. Cooley) show that self is often pre-verbal and that exclusion of an individual from her/his surroundings takes place also with the aid of the body and emotions tightly connected with functioning of self. The analysis of interactions between humans and animals provides us with much methodological and theoretical inspiration. Those researches and analyses obviously face a problem of “anthropomorphization of human behaviour”, which is of frequent occurrence both among researchers and ordinary people. New sociological sub-discipline called the sociology of human - non-human animals relationships adds a lot of new threads to the abovementioned deliberations on conditions of constructing self.

Opis:
The authors attempt to explore the positives of cross-cultural interactions in multinational companies (MNCs). The prior research has generally accentuated the problems in the relationship among foreigners and less effort has been made so far to examine the benefits of multiculturalism. Therefore, there is a need to fill in the gap identified above by providing the evidence that professional contacts with representatives from a different national cultural background may be very useful for both MNCs and individuals who are employed by them. The results of the authors' study suggest that the following major positives can be obtained due to cultural diversity, namely: 1) personal growth; 2) flexibility in working styles; 3) enhanced learning; 4) more perspectives, self-reflection; 5) individual benefits; 6) more effective communication; and finally 7) confrontation with stereotypes.

Informal Taxonomies of Charges of Correctional Facilities and Shelters for Juveniles Created by Personnel in the Context of Social Rehabilitation Interactions. Sociological Analysis of the Phenomenon

Opis:
The main aim of this paper is to draw attention to the phenomenon of creating informal taxonomies by the personnel of correctional institutions (ZP) and shelters for juveniles (SdN) towards charges in relation to social rehabilitation work. It discusses both the mechanisms of creating such classifications as well as the consequences associated with them. The article defends the thesis that this type of stigmatization is on the one hand a natural phenomenon, because it is based on the mechanism of analogy, to a large extent unconscious, typical for educational institutions, but on the other hand dependent in its form on the adopted style of institution management. It also discusses the problem of multiple consequences of taxonomies. The results presented in this paper are based on seven years qualitative research carried out in social rehabilitation centers of all types throughout Poland. The research applied the principle of triangulation in the scope of methods, the researcher and data. To analyze empirical data an interpretative paradigm was used, which enables to see the interactive nature of the formation of negative emotions and emotional work.
