With the rapid development of the wind energy industry, there is an increasing concern about operation safety and reliability of high-strength wind turbine bolts. The aim of this paper is to monitor the strain change around the cracks in wind turbine bolts by means of fiber Bragg grating (FBG) sensors for crack detection. Firstly, the strain distributions of wind turbine bolts’ cracks with different locations and angles in the service condition are simulated using finite element analysis (FEA). Then, three-point grating string FBG sensors were pasted on the surface of wind turbine bolts with fatigue cracks to monitor the strain changes around the cracks in real time. By analysing the monitored strain data elaborately, the location of the crack on the bolt surface was successfully detected by identifying the location of the maximum strain detected by FBG sensors. In addition, the strain distributions in the vicinity area of the crack at different angles (0◦, 45◦and 90◦) were also monitored and analysed in depth. The different types of crack angles could be distinguished based on of different strain distribution of the vicinity of the crack tip at different angles. The experimental results show that the FBG sensing technology has a high degree of sensitivity and accuracy in crack detection of high-strength wind turbine bolts.

Looseness of high-strength wind turbine bolts is one of the main types of mechanical failure that threaten the quality and safety of wind turbines, and how to non-destructively detect bolt loosening is essential to accurate assessment of operational reliability of wind turbine structures. Therefore, to address the issue of looseness detection of high-strength wind turbine bolts, this paper proposes a non-destructive detection method based on digital image correlation (DIC). Firstly, the mathematical relationships between the in-plane displacement component of the bolt’s nut surface, the bolt’s preload force loss and the bolt loosening angle are both deduced theoretically. Then, experimental measurements are respectively conducted with DIC with different small bolt loosening angles. The results show that the bolt loosening angle detection method based on DIC has a detection accuracy of over 95%, and the bolt’s preload force loss evaluated by the deduced relationship has a good agreement with the empirical value. Therefore, the proposed DIC-based bolt loosening angle detection method can meet the requirements of engineering inspection, and can achieve quantitative assessment of preload forces loss of wind turbine bolt.

The normal mode solution for the form function and target strength (TS) of a solid-filled spherical shell is derived. The calculation results of the spherical shell’s acoustic TS are in good agreement with the results of the finite element method (FEM). Based on these normal mode solutions, the influences of parameters such as the material, radius, and thickness of the inner and outer shells on the TS of a solid-filled spherical shell are analyzed. An underwater spherical shell scatterer is designed, which uses room temperature vulcanized (RTV) silicone rubber as a solid filling material and does not contain a suspension structure inside. The scatterer has a good TS enhancement effect.

An active inerter-based suspension with acceleration feedback control is proposed in this paper, the time delay generated in the controllers and actuators is considered, which constitutes the time-delayed active inerter-based (TDA-IB) suspension. The dynamic equation of the TDA-IB suspension is established and is a neutral type of delay differential equation (NDDE) in which the time delay exists in the highestorder derivative. The stability analysis is conducted by calculating the number of unstable characteristic roots based on the definite integral stability method, the stable and unstable regions are determined. The effect of time delay and feedback gain on the dynamic performance of the TDA-IB suspension under harmonic, random, and shock excitations is studied in detail and compared with the parallel-connected inerterbased (PC-IB) and traditional suspensions. The results show that the TDA-IB suspension is asymptotically stable for smaller feedback gain and time delay, through increasing the feedback gain, the stable regions shrink, and a smaller time delay could cause the system to become unstable. Furthermore, the time delay could regulate the resonance peak around the unsprung mass natural frequency and generate multiple high-frequency resonance peaks. If the time delay is chosen appropriately and falls into the stable range, the TDA-IB suspension could improve the dynamic performance for the suspension stroke and dynamic tire load while having a deterioration for the vehicle body acceleration compared with the PC-IB and traditional suspensions.

Insulin resistance (IR) plays a critical role in metabolic syndrome (MS). Previous studies have demonstrated that activated ROCK is increased in MS patients. However, the effect of Rho-kinase (ROCK) on IR has not been definitely determined. Thus, the aims of the present study were to determine whether ROCK activation induces IR or affects myocardial structure and function, as well as the possible mechanisms underlying this process. Wistar rats fed high fat, high glucose and high salt diet sewed as model of MS and we used transmission electron microscopy, echocardiogram technology, and terminal deoxynucleotidyl transferase-mediated DNA nick-end labeling staining to identify any myocardial damage. The protein levels of MYPT-1 (characteristic of ROCK activation), IRS-1 and AKT were analyzed by immunohistochemistry and Western blotting. In hearts from MS rats, we found increased protein levels of phospho-MYPT-1 and phospho-IRS-1 (Ser307) and decreased phospho-AKT compared to levels in normal rats. In conclusion, the results suggest that ROCK-mediated IR is involved in the development of myocardial impairments in MS rats and that this effect is mediated probably via the IRS-1/PI3-kinase/AKT pathway.

Peptidylarginine deiminase (PAD) catalyzes the post-translational modification of protein through the conversion of arginine to citrulline in the presence of calcium ions. Human, similar to rodents, has four isoforms of PAD (type I, II, III and IV/V), each of which is distinct in substrate specificity and tissue specific expression. In our large-scale sequencing project, we identified a new human PAD cDNA from a human fetal brain cDNA library. The putative protein encoded by this cDNA is designated hPADVI. Expression analysis of hPADVI showed that it is mainly expressed in adult human ovary and peripheral blood leukocytes. We conclude that hPADVI may be orthologous to mouse ePAD, basing on sequence comparison, chromosome localization and exon-intron structure analysis. PAD-mediated deimination of epithelial cell keratin resulting in cytoskeletal remodeling suggests a possible role for hPADVI in cytoskeletal reorganization in the egg and in early embryo development. This study describes a new important member of the human PAD family.

Phage ΦC31 integrase is a potential vector for the insertion of therapeutic genes into specific sites in the human genome. To understand the mechanism involved in ΦC31 integrase-mediated recombination, it is important to understand the interaction between the integrase and cellular proteins. Using a yeast two-hybrid system with pLexA-ΦC31 integrase as bait, we screened a pB42AD human fetal brain cDNA library for potential interacting cellular proteins. From the 106 independent clones that were screened, 11 potential interacting clones were isolated, of which one encoded C-terminal fragment of Sp100. The interaction between Sp100 and ΦC31 integrase was further confirmed by yeast mating and co-immunoprecipitation assays. The hybridization between a ΦC31 integrase peptide array and an HEK293 cell extract revealed that residues 81RILN84 in the N-terminus of ΦC31 integrase are responsible for the interaction with Sp100. Knocking down endogenous Sp100 with Sp100-specific siRNA increased ΦC31 integrase-mediated recombination but did not impact reporter gene expression. Therefore, endogenous Sp100 may interact with ΦC31 integrase and inhibit the efficiency of ΦC31 integrase-mediated recombination.
