Activity of hydrolases during the third and fourth larval stage of Anisakis simplex was identified by applying the API ZYM test method. In A. simplex larvae the activity of phosphatases was high, particularly that of acid phosphatase (40 nmol/mg-'). Among esterases lack of activity of lipase (Cj 4) is worth noticing while the activity of esterases (C4) and (Cg) was high. The activity of those later two enzymes was higher in L larvae than in Ly larvae. The highest activity in the subclass of glucosidases was recorded for B-fucosidase and N-acetyl-B-glucosaminidase. A higher activity in L3 larvae than in Ly larvae was recorded for: B-glucuronidase and N-acetyl-B-glucosaminidase (2-fold) and B-fucosidase (3-fold). Differently the activity of B-galactosidase and B-glucosidase was higher in L4 larvae than in L3 larvae. The tests did not show activity of o-galactosidase, 8-glucosidase and amannosidase on both larval forms.

Hydrolase activity of (enzymograms, biotypes) in Geotrichum candidum, one of the poorly described pathogenic fungi, was studied 81 strains were isolated from oral cavity and faeces of patients with gastrointestinal tract disorders. Axenic strains were differentiated with API 20C Aux and API ZYM tests. Then, enzymograms and biotypes were determined for all strains based on the activity of 19 hydrolases. High variability of enzymograms (17 different types) was found. The highest activity was noted in case of: e2-alkaline phosphatase, e6-lecucine arylamidase, e11-acid phosphatase. E5-lipase. e7-valine arylamidase, e12 - naphtol-AS-BI-phosphohydrolase and e17-β-glucosidase were used for biotyping procedures. Our own system of biotyping of 81 strains of G. candidum was based on the mathematical binominal distribution formula (1 : 4 : 6 : 4: 1) – all; ,,+”;one „-”, three „+”; two „-”,two „+”; three ,,-”,one ,,+”; all „-”. We have found: A (11.1 ± 3.5%), B1 (6.17 ± 2.67%). B2 (1.23 ± 1.22%), B4 (4.94 ± 2.41%), C1 (1.23 ±1.22%), C3 (63.0 ± 5.4%), D2 (9.88 ±3.31%), D3 (2.47 ± 1.72%). Among all strains from 8 various biotypes of G. candidum.

Similar patterns of hydrolases were observed in three species representing two genera of entomopathogenic nematodes: Steinernema affinis, S. feltiae, and Heterorhabditis zealandica. The same enzymes were present in the studied nematodes but they differed in the level of activity of individual sub-classes of enzymes. A higher activity of esterases and proteolytic enzymes could be observed for H. zealandica than for S. affinis and S. feltiae. On the other hand, the activity of glycosidases in steinernematids was generally much higher than in H. Zealandica.

Enzymes capable of converting l-asparagine to l-aspartate can be classified as bacterial-type or plant-type l-asparaginases. Bacterial-type l-asparaginases are further divided into subtypes I and II, defined by their intra-/extra-cellular localization, substrate affinity, and oligomeric form. Plant-type l-asparaginases are evolutionarily and structurally distinct from the bacterial-type enzymes. They function as potassium-dependent or -independent Ntn-hydrolases, similar to the well characterized aspartylglucosaminidases with (αβ)2 oligomeric structure. The review discusses the structural aspects of both types of l-asparaginases and highlights some peculiarities of their catalytic mechanisms. The bacterial-type enzymes are believed to have a disordered active site which gets properly organized on substrate binding. The plant-type enzymes, which are more active as isoaspartyl aminopeptidases, pose a chemical challenge common to other Ntn-hydrolases, which is how an N-terminal nucleophile can activate itself or cleave its own α-amide bond before the activation is even possible. The K+-independent plant-type l-asparaginases show an unusual sodium coordination by main-chain carbonyl groups and have a key arginine residue which by sensing the arrangement at the oligomeric (αβ)-(αβ) interface is able to discriminate among substrates presented for hydrolysis.

Nudix hydrolases are a family of proteins defined by a conserved amino-acid sequence GX5-EX7REUXEEXGU, where U is a hydrophobic residue. These enzymes are widely distributed among all classes of organisms and catalyze, with varying degrees of substrate specificity, the hydrolysis of a variety of nucleoside diphosphate derivatives: nucleoside di- and triphosphates and their oxidized forms, dinucleoside polyphosphates, nucleotide sugars, NADH, coenzyme A and the mRNA cap. Nudix proteins are postulated to control the cellular concentration of these compounds. The genome of the model plant Arabidopsis thaliana contains 29 genes coding for putative Nudix hydrolases. Recently, several Arabidopsis Nudix genes have been cloned and their products characterized. This review summarizes current knowledge on these plant enzymes and discusses their possible cellular functions.
