Cirrhosis leads to an inability of the liver to perform its biochemical functions. It can also lead to hepatocellular carcinoma in which, as we showed lately, arginase isoenzyme pattern changes. The present work presents our results on arginase isoenzymes and their possible role in liver cirrhosis. The study was performed on tissues obtained during liver transplantation from 60 patients with liver cirrhosis, and on samples of histologically normal liver (control) from 40 patients with benign or colorectal cancer liver metastases removed during surgery, 6-7 cm from the tumor border. Arginase isoenzymes AI (so-called liver-type arginase) and AII (called extrahepatic arginase) were identified by Western blotting and isolated by ion-exchange chromatography. Their expression on mRNA level was studied by RT-PCR. A significant decrease in arginase activity, dependent of the liver clinical stage, was observed in cirrhotic tissue. Arginase AI activity and its mRNA level were significantly decreased in cirrhotic liver, whereas the activity and expression of arginase AII were concurrently raised, as compared to normal liver. Since arginase AI is a key enzyme of the urea cycle, whereas arginase AII most probably takes part in the biosynthesis of ornithine and polyamines, the defective ammonia inactivation and increased collagen biosynthesis observed in cirrhotic liver may be related to the changes in arginase AI and AII levels, respectively.

The usefulness of mutagenic treatment to enlarge isozymic variability of barley and the use of induced mutants for genetic analysis were evaluated. N-methyl-N-nitroso urea, sodium azide and gamma rays were employed as mutagenic agents. Electrophoretic assays of 3848 M₂ seedlings obtained by chemical mutagenic treatment of the spring barley cultivars Dema, Aramir, Bielik and 3100 M₂ seedlings obtained by physical mutagenic treatment of the cv. Dema revealed 70 isozymic mutants, which represent 30 separate mutants in 25 M₁ plants. Most of mutations (27) were induced by chemical mutagen at polymorphic esterase loci. The occurrence of induced mutants at monomorphic loci, Got2 and Lap2, made it possible to perform genetic analysis of those loci in barley including mapping respective genes within chromosomes.

Evolutionary processes lead to the survival of individuals best adapted to local environment. This gives rise to allele polymorphism and genetic diversity of populations. Isoenzyme proteins, which are the product of gene expression, are an effective tool for tracking these changes. On the other hand, the reproductive potential of a given population can be assessed based on its ability to produce viable and efficiently germinating seeds. The present results combine molecular analyses of isoenzyme proteins with anatomical and morphological studies of Scots pine seeds (Pinus sylvestris L.). The study was conducted in 6 populations that are characteristic of this species occurrence range in the country. The results confirm the correlation between seed weight and embryo size. They also show a population from northeastern Poland had a higher effective number of alleles and seed with lower germinative energy and capacity. There was genetic homogeneity in all except for the population from Woziwoda, which was significantly different based on the Fst test. The genetic characteristics of Scots pine from Woziwoda may be associated with the lower levels of rainfall that occur there during the growing season. The results improve our knowledge of Scots pine variability and contribute to the discussion of the impact of local environment on genetic variability.

CK2 is a pleiotropic constitutively active serine/threonine protein kinase composed of two catalytic α- and two regulatory β-subunits, whose regulation is still not well understood. It seems to play an essential role in regulation of many cellular processes. Four active forms of CK2, composed of αα'ββ', α2ββ', α'2ββ', and a free α'-subunit were isolated from wild-type yeast and strains containing a single deletion of the catalytic subunit. Each species exhibits properties typical for CK2, but they differ in substrate specificity and sensitivity to inhibitors. This suggests that each CK2 isomer may regulate different process or may differ in the way of its regulation.

Seed protein profile and esterase isoenzme were studied in ten accessions of lemongrass [Cymbopogon flexuosus] collected from northeastern India belonging to two major chemotypes – citral and geraniol rich essential oil. Total four esterase isozyme bands were ob55 Vol. 57 No. 3 2011 Seed protein and esterase isozyme profile among the accession of lemongrass served with Rm values in the range of 0.394 to 0.798. Among them one with Rm value 0.747 was unique as it was consistently present in all the accessions irrespective of citral or geraniol rich chemotype. For seed protein, SDS-PAGE analysis of seven accessions (RLJ-TC-1, RLJ-TC-4, RLJ-TC-5, RLJ-TC-8, RLJ-TC-9, RLJ-TC-10 and OD-19) revealed a total of 13 bands among the accessions ranging in the size from 21.5 to 92.0 kd. Six citral rich accessions exhibits very similar seed protein profile with 10 to 11 protein band each. However, the geraniol rich chemo-type RLJ-TC-8 exhibit different profile with only six high molecular proteins. Four seed proteins with molecular weight 92.0, 86.0, 80.4 and 61.6 kd were consistently found in all the chemotypes irrespective of citral or geraniol rich and can be considered as marker for the species. Although esterase isozymes exhibited low polymorphism, yet close similarity of isozyme and seed protein profile can be considered as evidence of genetic homogeneity among the accession of lemongrass.
Badano profil białkowy nasion i izoenzymów esterazy w próbkach palczatki pogiętej [Cymbopogon flexuosus (Nexx ex Steud.) Wats] pochodzących z dziesięciu stanowisk, zebranych w północno-wschodniej części Indii, należących do dwóch głównych chemotypów ze względu na zawartości olejków eterycznych: typu cytralowego i geraniolowego. Współczynnik ruchliwości elektroforetycznej Rm badanych prób wahał się w granicach 0,394– 0,798. Spośród nich współczynnik RM o wartości 0,747 wyróżniał się wśród pozostałych i występował na wszystkich analizowanych stanowiskach niezależnie od charakteru chemotypu. Rozdział elektroforetyczny SDS-PAGE profilu białkowego nasion roślin pochodzących z siedmiu stanowisk (RLJ-TC-1, RLJ-TC-4, RLJ-TC-5, RLJ-TC-8, RLJ-TC-9, RLJ-TC-10 i OD+19) charakteryzował specyficzny układ 13 prążków w zakresie wielkości od 21,5 do 92 kD. U roślin należących do chemotypu cytralowego (6 osobników) stwierdzono podobny układ 10–11 prążków. Jednakże w chemotypie bogatym w geraniol RLJ-TC-8 stwierdzono odmienny profil odpowiadający sześciu wysokocząsteczkowym białkom. W przypadku wszystkich badanych chemotypów obserwowano powtarzający się układ 4 prążków dla białek o wielkościach odpowiednio: 92,0, 86,0, 80,4, 61,6 kd, co pozwala uznać go za marker charakterystyczny dla badanych roślin. Pomimo występowania niskiego stopnia polimorfizmu izoenzymów esterazy, przy jednoczesnej wysokiej zgodności pomiędzy wynikami rozdziału elektroforetycznego izoenzymów i profilu białkowego nasion, można wnioskować o genetycznej homogenności badanych stanowisk palczatki pogiętej.

Identity of reproductive material is essential in artificial forest regeneration. Legal regulations cannot guarantee proof of identity. State and private organizations have been cooperating to develop a seed certification scheme in forestry. Representative samples of forest reproductive material are taken at any production stage starting at the time of seed collection through to plant delivery as well as at time of changing ownership.The first sample at the time of seed harvest is used to determine the maximum potential yield of living germinants from the total collection. Furhtermore this and the other samples are tested randomly at a determined level of intensity or in case of doubt by Isozyme/DNA methods. Sampling techniques have been tested and described in a handbook. The biochemical-genetic tests are being standardized for all major commercial species. Forest reproductive material having been sampled in the described way will be certified "of provable identity" by a neutral agency. The Scheme will be open to anyone complying with the rules laid down. Larger forest owners have confirmed to preferably purchase "forest plants of provable identity".

This study investigates the rates of grafting and planting errors that occur in seed orchards, and evaluates their effects on the genetic diversity and relatedness of genotypes. The study used three clonal Scots pine seed orchards of differing ages and clonal composition located in the forest districts of Susz, Pniewy and Zdrojowa Góra, Poland. Maximum breeding ability within a seed orchard requires isolated from external sources of pollen, and have no alien genotypes inside the orchard. We used 13 isoenzyme markers to determine the genotypic identity of ramets and compared the number of genotypes of the actual current ramet population (W1) with genotypes comprising the originally intended plus trees (designated as W0) to estimate the genotypic assignment error rate per orchard. For both W0 and W1, we calculated the effective number of clones and the relative effective number of clones. Ramet assignment errors were detected in all three seed orchards. Gnotypic errors ranged from 5.8% to 37.7% across orchards. A total of 46 alleles were found, with the mean number of alleles per locus ranging from 2.77 to 3.23. At individual loci, the level of observed heterozygosity was variable. Alien genotypes had negligible effects on seed orchard heterozygosity. The Fst values between seed orchards amounted to 0.6% between Susz and Pniewy and 1.1% between Susz and Zdrojowa Góra. The effect on genetic variation of ramet assignment errors was small and influenced genetic diversity only in the case of the Susz seed orchard. However, our results suggest that negative effects of alien genotypes can occur on breeding value of seeds from analysed seeds orchards.
