Opis:
Tomato is an economically important vegetable crop which is attacked heavily by insect pests leading to reduction of yield and quality of the fruits. Field experiments were carried out to investigate the dissipation of methomyl (a common insecticide) used mainly on tomato fruits. LC-MS/MS coupled with the QuEChERS method were used for the determination of methomyl. The results showed that the recovery using matrix-matched standards ranged from 87.8 to 101.3%, with relative standard deviation of 2.5 to 7.5%. Kinetics equation, Log R = log R0 – 0.434 Kt, was used to calculate the rate of degradation in tomato, soil and water. Residue half-life calculated using kinetic rate ranged from 1.95 to 1.63 days in tomato and soil, respectively. From the results it was concluded that tomato fruits can be safely harvested for consumption after 15 days of application based on estimated preharvest interval (PHI). It is advisable to re-estimate the PHI regularly owing to data from the EU and Codex.

Opis:
The cowpea aphid (Aphis craccivora Koch) is considered a serious insect pest attacking several crops. We carried out biochemical studies to elucidate the role of the metabolising enzymes in conferring resistance to thiamethoxam, in two strains (resistant and susceptible) of the cowpea aphid. Bioassay experiments showed that the thiamethoxam selected strain developed a 48 fold resistance after consecutive selection with thiamethoxam for 12 generations. This resistant strain also exhibited cross-resistance to the tested carbamates; pirimicarb and carbosulfan, organophosphorus (malathion, fenitrothion, and chlorpyrifos-methyl), and the neonicotinoid (acetamiprid). Synergism studies have indicated that S,S,S-tributyl phosphorotrithioate (DEF), a known inhibitor for esterases, increased thiamethoxam toxicity 5.58 times in the resistant strain compared with the susceptible strain. Moreover, the biochemical determination revealed that carboxylestersae activity was 30 times greater in the resistant strain than in the susceptible strain. In addition, the enzyme activity of glutathione S-transferase (GST) and mixed function oxidases (mfo) increased only in the resistant strain 3.7 and 2.7 times, respectively, in relation to the susceptible (the control). Generally, our results suggest that the higher activity of the detoxifying enzymes, particularly carboxylesterase, in the resistant strain of the cowpea aphid, apparently have a significant role in endowing resistance to thiamethoxam, although additional mechanisms may contribute.

The primary purpose of this work was the initiation and optimization of shoot cultures of different Vitis vinifera L. cultivars: cv. Chardonnay, cv. Hibernal, cv. Riesling, cv. Johanniter, cv. Solaris, cv. Cabernet Cortis, and cv. Regent. Cultures were maintained on 30-day growth cycles using two media, Murashige and Skoog (MS) and Schenk and Hildebrandt (SH), with various concentrations of plant growth regulators. Tested media (‘W1’–‘W4’) contained varying concentrations of 6-benzylaminopurine (BA) in addition to indole-3-butyric acid (IBA) and 1-naphthaleneacetic acid (NAA). High performance liquid chromatography coupled with mass spectrometry (UPLC–MS) was used for metabolomic profiling. In all tested extracts, 45 compounds were identified (6 amino acids, 4 phenolic acids, 13 flavan-3-ols, 3 flavonols, and 19 stilbenoids). Principal component analysis (PCA) was performed to assess the influence of the genotype and medium on metabolic content. PCA showed that metabolic content was mainly influenced by genotype and to a lesser extent by medium composition. MS media variants induced the amino acid, procyanidin, and flavan-3-ol production. In addition, the antioxidant potential and anti-tyrosinase activity was measured spectrophotometrically. The studies on antioxidant activity clearly reveal very high efficiency in reducing free radicals in the tested extracts. The strongest tyrosinase inhibition capacity was proved for shoots cv. Hibernal cultured in SH medium and supplemented with NAA, with an inhibition of 17.50%. These studies show that in vitro cultures of V. vinifera cvs. can be proposed as an alternative source of plant material that can be potentially used in cosmetic industry.

Opis:
Integration of the remote sensing techniques including the thermal emission and reflection radiometer (ASTER) thermal infrared (TIR) data, and the silica index (SI) algorithms have enabled the discovery of a previously unknown white sand deposit in the El-Harra region of the Bahariya Oasis. The physical, chemical, and mineralogical characterization of El-Harra white sand was achieved using the binocular microscope, X-Ray Diffraction (XRD), X-Ray Fluorescence spectrometry (XRF) and Scanning Electron Microscopy (SEM). The sample was physically beneficiated using attrition scrubbing, shacking table concentration and magnetic separation while its heavy mineral content was conducted via heavy liquid separation. The chemical and grain size analysis before and after the physical treatment pointed to clear enhancing in both the chemical and grain size specifications, where the oxide ratios of SiO2, Al2O3, Fe2O3 and CaO moved from 96.87, 0.82, 0.13 and 1.27% to 99.53, 0.05, 0.05 and 0.13%, respectively. On the other hand, the ratio of grain size (–0.5/+0.125 mm) was raised from 79.48% to 82.04% from the total sample weight. Comparing these data with their corresponding in both the British and American Standard Specifications of White Sand confirmed the validity of the physical beneficiation in raising the grade of the investigated white sand from low valuable ore to the globally highest one which means higher marketing prices and the availability to use the treated white sand in a wide spectrum of technological and strategic applications. Furthermore, the heavy minerals content and species is another add value, where their content was assigned as 0.3 mass% from the head sample including ilmenite, rutile, zircon, leucoxene and green silicates. This simply means each 1000 tons of physically beneficiated white sand yield 3 tons of the heavy minerals.

COVID-19 vaccines are crucial to control the pandemic and avoid COVID-19 severe infections. The rapid evolution of COVID-19 variants such as B.1.1.529 is alarming, especially with the gradual decrease in serum antibody levels in vaccinated individuals. Middle Eastern countries were less likely to accept the initial doses of vaccines. This study was directed to determine COVID-19 vaccine booster acceptance and its associated factors in the general population in the MENA region to attain public herd immunity. We conducted an online survey in five countries (Egypt, Iraq, Palestine, Saudi Arabia, and Sudan) in November and December 2021. The questionnaire included self-reported information about the vaccine type, side effects, fear level, and several demographic factors. Kruskal-Wallis ANOVA was used to associate the fear level with the type of COVID-19 vaccine. Logistic regression was performed to confirm the results and reported as odds ratios (ORs) and 95% confidence intervals. The final analysis included 3041 fully vaccinated participants. Overall, 60.2% of the respondents reported willingness to receive the COVID-19 booster dose, while 20.4% were hesitant. Safety uncertainties and opinions that the booster dose is not necessary were the primary reasons for refusing the booster dose. The willingness to receive the booster dose was in a triangular relationship with the side effects of first and second doses and the fear (p < 0.0001). Females, individuals with normal body mass index, history of COVID-19 infection, and influenza-unvaccinated individuals were significantly associated with declining the booster dose. Higher fear levels were observed in females, rural citizens, and chronic and immunosuppressed patients. Our results suggest that vaccine hesitancy and fear in several highlighted groups continue to be challenges for healthcare providers, necessitating public health intervention, prioritizing the need for targeted awareness campaigns, and facilitating the spread of evidence-based scientific communication.

Background and Objectives: The efficacy of deep brain stimulation of the anterior nucleus of the thalamus (ANT DBS) in patients with drug-resistant epilepsy (DRE) was demonstrated in the double-blind Stimulation of the Anterior Nucleus of the Thalamus for Epilepsy randomized controlled trial. The Medtronic Registry for Epilepsy (MORE) aims to understand the safety and longer-term effectiveness of ANT DBS therapy in routine clinical practice. Methods: MORE is an observational registry collecting prospective and retrospective clinical data. Participants were at least 18 years old, with focal DRE recruited across 25 centers from 13 countries. They were followed for at least 2 years in terms of seizure frequency (SF), responder rate (RR), health-related quality of life (Quality of Life in Epilepsy Inventory 31), depression, and safety outcomes. Results: Of the 191 patients recruited, 170 (mean [SD] age of 35.6 [10.7] years, 43% female) were implanted with DBS therapy and met all eligibility criteria. At baseline, 38% of patients reported cognitive impairment. The median monthly SF decreased by 33.1% from 15.8 at baseline to 8.8 at 2 years (p < 0.0001) with 32.3% RR. In the subgroup of 47 patients who completed 5 years of follow-up, the median monthly SF decreased by 55.1% from 16 at baseline to 7.9 at 5 years (p < 0.0001) with 53.2% RR. High-volume centers (>10 implantations) had 42.8% reduction in median monthly SF by 2 years in comparison with 25.8% in low-volume center. In patients with cognitive impairment, the reduction in median monthly SF was 26.0% by 2 years compared with 36.1% in patients without cognitive impairment. The most frequently reported adverse events were changes (e.g., increased frequency/severity) in seizure (16%), memory impairment (patient-reported complaint, 15%), depressive mood (patient-reported complaint, 13%), and epilepsy (12%). One definite sudden unexpected death in epilepsy case was reported.
