Objectives. To determine whether the degree of hamstring muscle extensibility influences the sagittal spinal curvatures in standing and maximal trunk flexion on young athletes. Methods. Forty-two young elite kayakers (mean age: 15.09 ± 0.63 years) were recruited. Thoracic and lumbar curvatures and pelvic position were evaluated with a Spinal Mouse system in standing position and maximal trunk flexion with knees extended (toe-touch test) and flexed. Hamstring muscle extensibility was determined by passive straight leg raise test (PSLR). The sample was divided into two groups with regard to straight leg raise angle (PSLR < 80º, n=20, and PSLR ≥ 80º, n=20). Results. Subjects with lower extensibility presented higher thoracic angle and a more posterior pelvic tilt in maximal trunk flexion. However, no significant differences were found between both groups when standing. The lumbar curve was not affected by hamstring extensibility. Conclusions. Lower hamstring extensibility is related to increased thoracic curve and more posterior pelvic tilt when maximal trunk flexion is performed, which can overload the spine. A systematic stretching programme to improve hamstring extensibility should be incorporated into training activities.

Opis:
Objective: to evaluate the efficacy of a 32-week school physical education programme on low-back pain in elementary and secondary schoolchildren. Material and methods: Forty-one elementary school children (fifth-grade, mean age of 10.27 ± 0.31 years) and 43 secondary school adolescents (twograde, mean age of 13.46 ± 0.68 years) were assigned to the control (n = 40) or intervention group (n = 44). The intervention subjects were involved in an organised physical education programme including hamstrings stretching, endurance strength of the abdominal and lumbar muscles, and pelvic tilt during the two-weekly school physical education classes over 32 weeks. The control group was not subjected to the organized programme. Low back pain was registered and pain intensity was recorded using the Visual Analogue Scale. Results: The experimental group showed a statistically significant decrease of low back pain frequency while the control group evidenced an increase. For pain intensity no significant differences were found. Conclusion: The children and adolescents who were subjected to the school physical education programme showed a reduction of low back pain frequency, while a tendency toward the rising frequency of low back pain was detected for the control subjects.

Opis:
The clinical manifestations of human Chagas disease are associated with several factors, including immunological alterations, in this regard, many studies propose that tissue damage might be more severe in the absence of immune regulatory mechanisms, other factors are the genetic background of host and parasite. Trypanosoma cruzi population is genetically, biochemistry and pathogenic diverse along the Latin-America continent and phylogenetic ally are divided into six intra-species lineages TcI-VI. The TcI lineage has a wide distribution with heterogeneous virulence and pathogenesis within strains. In Mexico, the main circulating lineage is TcI in human infections. We analyzed intracytoplasmic cytokines of unstimulated peripheral T lymphocytes, and the level of cytokines (IL-2, IL-4, IL-12, IL-10, IFN-γ and sIL-2R) in the serum of Mexican chagasic subjects. The population studied consisted of 15 asymptomatic individuals, 17 patients with chronic chagasic cardiopathy (CCC), 20 patients with cardiopathy but negative serology for T. cruzi, and 10 healthy subjects. The analysis of CD4+ cells revealed that CCC and asymptomatic patients have higher CD25+ and CD69 activation markers than controls. The Th1 subset (CD4+/IFN- γ +) was higher in CCC than in asymptomatic and control subjects, whereas Th2 subset was markedly high in asymptomatic subjects. Circulating cytokines were below level detection with the exception of IL-2 and sIL-2R. Infection with Mexican Trypanosoma cruzi strains in asymptomatic chagasic subjects have a tendency for a Th2 response with higher CD8+/IFN-γ T cells. In contrast, CCC patients have low levels of intracellular IFN- γ and IL-2 cytokines. In both groups circulating serum cytokines are below the detectable level.

There are two principal applications oftransgenic animals. Best known andmost advanced application is to use transgenic animals (called bioreactors ormolecular farms) for the production of various proteins or biopolymers of medical significance. The second application concerns efforts to improve the productivity traits of breeding animals. We have worked out methods which allow somatic cloning of mammals and gene knockout methods. These methods havebeen developing very rapidly in recent years and their efficiency will soon beimproved to the extent that they will become profitable. For the time being,DNA microinjection with all its disadvantages, remains the principal method ofproducing transgenic bioreactors. In this paper, the effectiveness of productionoftransgenic rabbits, goats and pigs with the use of WAP-Fuc gene construct ispresented.

From weak interactions to high stability : deciphering the streptavidin–biotin interaction through NMR and computational analysis

Understanding weak interactions in protein–ligand complexes is essential for advancing drug design. Here, we combine experimental and quantum mechanical approaches to study the streptavidin–biotin complex, one of the strongest interacting protein–ligand systems. Using a monomeric streptavidin mutant, we analyze 1H NMR chemical shift perturbations (CSPs) of biotin upon binding, identifying remarkable upfield shifts of up to −3.2 ppm. Quantum chemical calculations attribute these shifts primarily to aromatic ring currents, with additional contributions from charge transfer effects linked to weak interactions. The agreement between experimental and computed chemical shifts validated the X-ray structure as a reliable basis for detailed computational analyses. Energy decomposition analysis reveals that electrostatics dominate the biotin–streptavidin interaction, complemented by significant orbital and dispersion contributions. Notably, weak noncovalent interactions, such as CH···S, CH···π, and CH···HC contacts, driven by London dispersion forces, contribute ∼44% to the complex’s stability.

Opis:
Remote controlled laboratories had a great push during the COVID-19 pandemic. In fact, they were already out there but lacking in visibility. This external trigger pushed the academy to face a global challenge to start offering remote experiments more consistently and maturely. Instituto Superior Técnico (IST) has been offering several remote experiments since 2000 but with the need for an update due to technological aging. As such, the framework for remote experiments in education (FREE) was created based on new web technologies. In addition to the most diverse experiments that had already been developed, FREE includes two experiments that aimed at advanced-level physics students: the Langmuir probe and the electromagnetic (EM) cavity. Both allow users to configure the various parameters and to access the results in real time or check back later. All this access is done using a browser (on a PC or mobile phone) without the need to install additional software. The results of an experimental execution are stored in a database and are downloadable, allowing users to do various analyses and to determine the corresponding plasma density and temperature. In this paper, we will introduce how FREE was used in the implementation of both experiments and give an insight into their didactic approach, such as: (i) how to perform an experimental execution, (ii) the typical data set obtained with, and (iii) the corresponding analysis necessary for the user to retrieve information from it.

Synthetic and characterization results of a new family of Fe(III) compounds stabilized by a trianionic [CF 3 − ONO] 3 − pincer-type ligand are reported. The ligand possesses three negatively charged donors constrained to the meridional positions that provide su ffi cient electron density to stabilize high-valent metal complexes. Using the redox-insulated [CF 3 − ONO] 3 − , pentacoordinated square-pyramidal {[CF 3 − ONO]- FeCl 2 }{LiTHF 2 } 2 ( 3 ), dimeric μ -DME{[CF 3 − ONO]- FeDME} 2 ( 4 ), trigonal bipyramidal [CF 3 − ONO]Fe(bpy) ( 5 ), and octahedral [CF 3 − ONO]Fe(bpy)H 2 O( 5 · H 2 O ) complexes are synthesized. An interesting feature of the [CF 3 − ONO] 3 − pincer-type ligand is its ability to coordinate the metal center in both the more common meridional positions or occupying a face of a trigonal bipyramidal complex. The molecular structure of 3 contains structural features similar to those of a rare square-planar high-spin Fe(II) complex, and the important role of the counterions in stabilizing a square-plane is emphasized. SQUID magnetometry measurements of 3 reveal its high-spin character, and cyclic voltammetry measurements indicate high oxidation state species are unstable. However, all compounds can be reduced, and in particular 5 displays a reversible reduction event at − 2255 mV versus ferrocene (Fc + /Fc) that can be assigned to either the Fe I /Fe 0 couple or 2,2 ′ - bipyridine reduction.
