The paper presents a proposal for improving the supply company Mercedes - Benz Danuta i Ryszard Czach Sp. z o.o. The improvements carried out a pilot not used so far in the company ABC method. The results were used to make organizational improvements in Mercedes - Benz Danuta i Ryszard Czach Sp. z o.o. They may be inspired similar actions in other companies.

Opis:: In the article enterprise acting in the business of processing plastics was researched. In the enterprise is realizes mainly mass production. The main factor that decides about the reliability of applied technologies there are used machines. As a result of analysis a modernity level of sub-assemblies for two machines was determined (injections moulding machine and extrusion machine). Analysis of the machines effectiveness based on chosen TPM coefficients was also undertaken. It allowed the assessment of the working time of these machines.

Tytuł:: Ekspresja transportera ksenobiotyków BCRP w tkankach i komórkach zwierzęcych
Characteristic of xenobiotic transporter BCRP in vitro and in vivo.

Opis:: Transporter ksenobiotyków BCRP należy do rodziny ABC, które przemieszczają różne substancje z wykorzystaniem energii z hydrolizy ATP. Bierze ono udział w transporcie ksenobiotyków i przyczynia się do oporności komórek na leki. BCRP jest zlokalizowane zarówno w tkankach zdrowych jak i w komórkach nowotworowych np. rak jajnika, czy szpiczaka mnogiego.Wyizolowano RNA z tkanek myszy szczepu DBA2 obarczonych guzem czerniaka 891 i oznaczono poziom mRNA dla BCRP. Ekspresja BCRP zależy zarówno od tkanki, w której dochodzi do jego ekspresji, jak i od wieku guza. Uzyskano także BCRP w drożdżowym systemie ekspresyjnym. Przeprowadzono izolację a następnie solubilizację BCRP z użyciem detergentu. Oczyszczanie białka BCRP przeprowadzono metodą chromatografii powinowactwa. Wyniki testów aktywności ATPazowej BCRP w membranach drożdżowych z inhibitorem Na3VO4, oraz dwoma substratami, Hoechst 33342 i feoforbidem, ukazują spadek aktywności ATPazowej BCRP w obecności tych związków. Ponadto, przygotowano jednowarstwowe liposomy o (Ø= 100 nm) i wyznaczono ich punkt solubilizacji w detergencie. Zsyntezowano endonadtlenki β-karotenu i zbadano ich wpływ na komórki Caco-2. W niskich stężeniach pochodne te działają na komórki umiarkowanie cytotoksycznie, natomiast w wysokich stężeniach prawdopodobnie stymulują wzrost komórek.
Transporter of xenobiotics BCRP belongs to ABC family which use the energy from ATP hydrolisis to transport different substances. This protein participates in xenobiotics transport and it’s also responsible for multidrug resistance. BCRP is localized in normal tissues and in cancer cells e.g. ovarian cancer or multiple myeloma.RNA was isolated from the tissues of mouse strain DBA2 with melanoma tumor 891 and next the mRNA and PCR technique was used to determine the level of native BCRP in these tissues. Expression of BCRP depends both on the tissue in which it expresses and also on the age of the tumor. BCRP were also obtained in a yeast expression system. BCRP was isolated and solubilized with a detergent. Purification of BCRP protein was performed by affinity chromatography. The results of ATPase activity assay show a decrease in ATPase activity of BCRP in the presence of Na3VO4, Hoechst 33342 and pheophorbide a. Moreover, unilamellar liposomes were prepared (Ø = 100 nm) and their point of solubilization with detergent was determined. Endoperoxides of β-carotene were synthesized and examined their effect on Caco-2 cells. At low concentrations, these derivatives are moderate cytotoxic, whereas at high concentrations they may stimulate cell growth.

Opis:: W artykule przedstawiono istotę funkcjonowania łańcucha dostaw i zarządzania zapasami w sektorze leśno-drzewnym w Polsce. Badanym obiektem była celowo dobrana jednostka organizacyjna lasów państwowych położona w rejonie puszczy Augustowskiej. Zdefiniowano metody sterowania zapasami w łańcuchu dostaw oraz usystematyzowano sposoby składowania drewna. Następnie przeprowadzono badania empiryczne w zakresie klasyfikacji zapasów w wybranym nadleśnictwie. Dzięki klasyfikacji zapasów metodą ABC określono, który asortyment przynosi największe przychody oraz wskazano sposoby usprawnienia działalności w obszarze zarządzania zapasami.
The thesis concerned the essence of stock management in forest and timber sector in Poland. The aim was focused on the acquisition and sale of wood raw material in a deliberately selected forest district, located in the region of the Augustow Primeval Forest. Defining the method of stock management in the supply chain, systematizing of wood storage methods. Then, empirical research was carried out on the classification of stocks in a selected forest district. Thanks to the classification of inventories using the ABC method, it was determined which assortment brings the highest revenues and ways to improve the activities in the area of inventory management.

Opis:: W artykule sprawdzono, jaki wpływ na średnie czasy (odległości) kompletacji ma umieszczenie towarów tego samego typu w wielu lokalizacjach magazynowych. Model uwzględniający wiele lokalizacji z tym samym towarem, zaproponowany przez Danielsa, Rummela i Schantza [1998], pozwala na znalezienie najkrótszej trasy w magazynie. W artykule omówiono problemy związane z praktycznym jego wdrożeniem. Skomentowano również propozycje Dmytrowa [2013; 2015] oraz Dmytrowa i Doszynia [2015] opierające się na takim wyborze pobieranych towarów, aby zmaksymalizować sumy wag przypisywanych lokalizacjom. W badaniach przyjęto, że wszystkie lokalizacje są równie atrakcyjne i skoncentrowano się na wyznaczeniu najkrótszej trasy przy założeniu, że magazynierzy poruszają się zgodnie z jedną z dwóch najczęściej wykorzystywanych w praktyce heurystyk: S-shape i return. Sprawdzono, jak na średni czas kompletacji zamówień wpływa zwiększanie liczby lokalizacji z tym samym towarem. Obliczenia wykonano za pomocą symulacji.
The paper examines how the number of locations with the same item affects the average distance covered by the picker. The model proposed by Daniels, Rummel and Schantz [1998] allows to search the shortest route. Unfortunately the problem is NP-hard. The problem of selection the location with items to be picked is treated by Dmytrów [2015] in a different way: here the three criteria are evaluated. In the paper, the one-block rectangular warehouse with S-shape and return routing is analyzed. The average distance traveled in one picking tour is counted for different storage policies. The main finding is that increasing the number of locations not always leads to the improvement of the order-picking process. The calculations were performed using simulations.

Opis:: Purpose: Appropriate product categorisation in warehouses is an issue facing most warehouses and distribution centres around the globe today. The ABC classification scheme aids in determining the most vital values at the warehouse. ABC classification reduces the effects of excess, end-of-life, or huge volumes of phase-out products. Design/methodology/approach: ABC classification is a method for classifying products according to their relevance. Products are divided into three categories using the ABC analysis, with category “A” items being the most important, category “B” as medium important and category “C” items being the least important for the distributor. This research model the ABC classification problem as a multi-knapsack and provides an example of commercial and noncommercial solver usage that enables distributors to categorise assortment according to the ABC classification. Findings: Proposed approach enables the distributor to arrive at the best possible outcomes. Research limitations/implications: The main limitation of this research is that it does not take into consideration constraints that show that some products cannot be quickly categorised or placed on the shelves because of the availability of storage equipment or warehouse personnel at the time of classification should be considered. Further research may be done on these issues. Practical implications: Performance comparisons between the proposed approach and the traditional ABC classification method provided by the distributors are conducted. Originality/value: The main contribution is the improvement of the classification method used in warehouses these days. The proposed approach allows assigning an optimal product mix to ABC categories.

Opis:: In the current situation where terrorism has become a dire and global concern, daily border crossings are fluctuating, further checks are being reinforced at border control procedures, biometric data can potentially contribute to a faster, more secure and feasible verification of people’s identity; thus tackling any related threats and constraints rising from the aforementioned matters.
