The aim of this work was to improve plating efficiency of sugar beet mesophyll protoplast cultures. Preliminary experiments showed that cultures of good quality, viable protoplasts were obtained in rich media based on the Kao and Michayluk formulation and with the calcium alginate as an embedding matrix. Nevertheless, in these cultures cell divisions were either not observed or very seldom confirming earlier reported recalcitrance of sugar beet protoplasts. The recalcitrant status of these cultures was reversed upon application of exogenous phytosulfokine (PSK)—a peptidyl plant growth factor. The highest effectiveness of PSK was observed at 100 nM concentration. Plating efficiencies obtained in the presence of PSK reached approximately 20% of the total cultured cells. The stimulatory effect of phytosulfokine was observed for all tested breeding stocks of sugar beet. Our data indicate that PSK is a powerful agent able to overcome recalcitrance of plant protoplast cultures.

Protoplast fusion was used to produce more than 500 symmetric interspecific somatic hybrids between wild Solanum species, which belong to the series Pinnatisecta, Etuberosa and S. tuberosum L. cultivars or potato breeding clones. The used genebank accessions of S. cardiophyllum and S. tarnii were resistant to Phytophthora infestans, while the accessions of S. cardiophyllum, S. tarnii, as well as of S. etuberosum, were also highly resistant to PVY (strains: PVYO, PVYN, PVYNTN, PVYC, PVYN Wilga). In most fusion combinations vigorous and genetically stable hybrid material has been selected. A number of interspecific somatic hybrids were fertile and could be backcrossed with cultivated potato. The use of detached leaf and tuber tests and mechanical inoculation, grafting and virus transmission by vectors, revealed that several somatic hybrids and backcross (BC) clones were resistant to foliage and tuber blight, as well as to PVY.

Opis:
The paper presents a new way of obtaining viable and very homogeneous cucumber protoplasts. Protoplasts from cells formed in the shoot tip meristem culture were isolated from suspension. Plasmid pBI121 was introduced using impulse electric field. Effectiveness of transformation process was determined by the transient expression of ß-glucuronidase (GUS) gene, controlled by promotor 35S. The activity of ß-glucuronidase enzyme as a product of GUS reporter gene was estimated by fluorimetric method (JEFFERSON 1987). Parameters of electroporation process were optimized. The transient expression of GUS gene was measured 24 h after electroporation. The highest effectiveness of transformation process was achieved using three electric impulses at the initial voltage of 250-350 V at 10-sec. intervals as a result of discharging a 140 µF capacitor and 50-70 µg × cm⁻³ plasmid DNA in the presence of 50 µg × cm⁻³ carrier DNA. The system presented is an effective method of exogenic DNA transfer, which is indicated by a high transient expression of the reporter gene. In comparison to Agrobacterium tumefaciens and A. rhizogenes, this alternative method of gene transfer can be used for obtaining transgenic cucumber plants.

Light microscopy with a bright field mode offers an easy and fast examination of plant specimen for carotenoid presence in its cells. Using basic techniques such as hand sectioned or squashed preparations, carotenoid-rich chromoplasts can be identified without applying any staining procedure and their localization within the cell, their shape and number can be assessed. More detailed information can be obtained by using Raman spectroscopy which is suitable for the analysis of carotenoids due to their unique Raman spectra and allows semiquantification of their contents. Raman imaging (mapping) can be additionally used to show the distribution of carotenoids within the sample. Raman spectra can be taken from extracted carotenoids but can be also obtained directly from plant tissues or cells as Raman measurements are nondestructive for the sample. Here we describe preparations of intact tissue samples, monolayer cell samples, isolated protoplasts as well as carotene crystals released from chromoplasts that are suitable for subsequent observations using light microscopy and for analysis using Raman spectroscopy.
