Opis:
Environmental and endogenous alkylating agents generate cytotoxic and mutagenic lesions in DNA. Exposure of prokaryotic cells to sublethal doses of DNA alkylating agents induces so called adaptive response (Ada response) involving the expression of a set of genes which allows the cells to tolerate the toxic and mutagenic action of such agents. The Ada response includes the expression of four genes: ada, alkA, alkB, and aidB. The product of ada gene, Ada protein, is an activator of transcription of all four genes. DNA bases damaged by alkylation are removed by distinct strategies. The most toxic lesion 3meA is removed by specific DNA glycosylase initiating base excising repair. The toxic and mutagenic O6meG is repaired directly by methyltransferases. 1meA and 3meC are corrected by AlkB DNA dioxygenase. The mechanisms of action of E. coli AlkB dioxygenase and its human homologs ABH2 and ABH3 are described in more details.

Opis:
Respiratory gas exchange in insects occurs via a branching tracheal system. The entrances to the air-filled tracheae are the spiracles, which are gate-like structures in the exoskeleton. The open or closed state of spiracles defines the three possible gas exchange patterns of insects. In resting insects, spiracles may open and close over time in a repeatable fashion that results in a discontinuous gas exchange (DGE) pattern characterized by periods of zero organism-to-environment gas exchange. Several adaptive hypotheses have been proposed to explain why insects engage in DGE, but none have attracted overwhelming support. We provide support for a previously untested hypothesis that posits that DGE minimizes the risk of infestation of the tracheal system by mites and other agents. Here, we analyze the respiratory patterns of 15 species of ground beetle (Carabidae), of which more than 40% of individuals harbored external mites. Compared with mite-free individuals, infested one’s engaged significantly more often in DGE. Mite-free individuals predominantly employed a cyclic or continuous gas exchange pattern, which did not include complete spiracle closure. Complete spiracle closure may prevent parasites from invading, clogging, or transferring pathogens to the tracheal system or from foraging on tissue not protected by thick chitinous layers.

W niniejszym artykule przeglądowym przedstawiono zagadnienie radiacyjnej odpowiedzi adaptacyjnej z punktu widzenia biofizycznego. Efekt ten – w dużym uproszczeniu – polega na wzmocnieniu mechanizmów naprawczych komórki oraz regulowaniu apoptozy i produkcji białek, w sytuacji napromienienia niską dawką lub mocą dawki promieniowania jonizującego. Jednakże efekt ten nie występuje zawsze, a jego powtarzalność eksperymentalna bywa częstokroć kwestionowana. Nie zmienia to faktu, że mechanizmy wywołujące odpowiedź adaptacyjną wymagają wciąż wielu badań, nie tylko radiobiologicznych, ale też fizycznych. W tym duchu powstało szereg modeli teoretycznych, a wybrane z nich zostały szczegółowo omówione w niniejszym artykule przeglądowym. Modele te stanowią dobry przykład możliwości współpracy na trójstyku biologii, fizyki i matematyki.
This review article presents the issue of radiation adaptive response from a biophysical point of view. This effect – to put it very simply – involves strengthening cell repair mechanisms and regulating apoptosis and protein production in the event of irradiation with a low dose or dose-rate of ionizing radiation. However, this effect does not always occur and its experimental repeatability is often questioned. This does not change the fact that the mechanisms causing the adaptive response still require a lot of research, not only radiobiological, but also physical. In this spirit, a number of theoretical models have been developed, and some of them are discussed in detail in this review article. These models are a good example of the possibilities of cooperation at the intersection of biology, physics and mathematics.

The paper concerns estimation of significance of differences of mutagenesis level between the wild-type strain (wt) and its derivatives which differ in DNA repair ability, namely alkA and alkB strain, devoided AlkA glycosylase and AlkB dioxygenase activity, respectively. The strains were analyzed for their ability to repair 1,N6-ethenoadenine (εA) - chloroacetaldehyde adduct to DNA. The analysis was done using classical statistical and pattern recognition methods. The obtained results confirmed that AlkB dioxygenase plays the most important role in εA repair in E. coli in the experimental modeling.

The practical application of time-variant reliability analysis is limited by its computationally expensive models which describe the structural system behavior. This paper presents a new adaptive PC-Kriging (APCK) approach to accurately and efficiently assess the time-variant reliabilities. Time interval is firstly discretized with a series of time instants and then the stochastic process is reconstructed by standard normal random variables and deterministic function of time. PC-Kriging (PCK) models are built at each time instant to predict the instantaneous responses of performance function. To improve the accuracy and efficiency, a new update strategy based on the integration of U- and H- learning functions is developed to refine the PCK models of instantaneous responses. One or two best samples are identified by the proposed learning criterion for updating the PCK models. Finally, Monte Carlo simulation (MCS) is used to estimate the time-variant reliability based on the updated PCK models. Four examples are used to validate the accuracy and efficiency of the proposed method.

Performances of underwater acoustic communication (UAC) systems are strongly related to specific propagation conditions of the underwater channel. Due to their large variability, there is a need for adaptive matching of the UAC systems signaling to the transmission properties of the channel. This requires a knowledge of instantaneous channel characteristics, in terms of the specific parameters of stochastic models. The wide-sense stationary uncorrelated scattering (WSSUS) assumption simplifies the estimation of terrestrial wireless channel transmission properties. Although UAC channels are hardly ever WSSUS, the rationale of such a stochastic modeling is that over a short period of time, and for a limited frequency range, this assumption is reasonably satisfied. The limits of application of the local-sense stationary uncorrelated scattering (LSSUS) model are determined by the stationary time and frequency. This paper presents the results of LSSUS model analysis for UAC channel measurements gathered by the former Research Department for Underwater Acoustics and Marine Geophysics (FWG), now part of the WTD71, during the SIMO experiment in the Bornholm Basin of the Baltic Sea.

Within the realm of machine learning, kernel meth-ods stand out as a prominent class of algorithms with widespreadapplications, including but not limited to classification, regres-sion, and identification tasks. Our paper addresses the chal-lenging problem of identifying the finite impulse response (FIR)of single-input single-output nonlinear systems under the in-fluence of perturbations and binary-valued measurements. Toovercome this challenge, we exploit two algorithms that leveragethe framework of reproducing kernel Hilbert spaces (RKHS) toaccurately identify the impulse response of the Proakis C chan-nel. Additionally, we introduce the application of these kernelmethods for estimating binary output data of nonlinear systems.We showcase the effectiveness of kernel adaptive filters in identi-fying nonlinear systems with binary output measurements, asdemonstrated through the experimental results presented in thisstudy.

Over the last few years, kernel adaptive filters have gained in importance as the kernel trick started to be used in classic linear adaptive filters in order to address various regression and time-series prediction issues in nonlinear environments.In this paper, we study a recursive method for identifying finite impulse response (FIR) nonlinear systems based on binary-value observation systems. We also apply the kernel trick to the recursive projection (RP) algorithm, yielding a novel recursive algorithm based on a positive definite kernel. For purposes, our approach is compared with the recursive projection (RP) algorithm in the process of identifying the parameters of two channels, with the first of them being a frequency-selective fading channel, called a broadband radio access network (BRAN B) channel, and the other being a a theoretical frequency-selective channel, known as the Macchi channel. Monte Carlo simulation results are presented to show the performance of the proposed algorithm.

The SOS system and SOS mutagenesis are frequently studied, or exploited to obtain an increase in mutagenicity of bacteria. Here a short survey is made of the phenomenon of SOS response with special attention to latest and less discussed data, especially the induction of the SOS system in response to cell starvation or mutation of certain genes and the role of inducible DNA polymerases.
