Opis:
Tea, Camellia sinensis L. is the most widely drunk beverage in the world. Comprehensive healing properties of this refreshing, mildly stimulating drink are known in traditional medicine dating back almost 5000 years. Tea healing and disease prevention mechanism is mostly based on antioxidant properties. Tea beverage is a rich source of polyphenols, especially catechins which are very powerful antioxidants. The most abundant catechins in tea beverage are epigallocatechin 3-gallate (EGCG), epigallocatechin (EGC), epicatechin 3-gallate (ECG), and epicatechin (EC). Many investigations i.e. population-based, screening, involving patients and also with animal or cell models, show many valuable pharmacological properties of tea. It is proved by numerous studies that drinking tea or taking tea extracts can prevent chronic diseases, including coronary heart disease and many types of cancer. It can also help with diabetes, hypertension and obesity and may contribute to inflammation reduce. Better understanding of tea healing properties could help reduce high costs of medical care, improve treatment of lifestyle diseases, and transform tea beverage into an evidence based functional food.

Opis:
Birds, like other vertebrates, rely on a robust antioxidant system to protect themselves against oxidative imbalance caused by energy-intensive activities such as flying. Such oxidative challenges may be especially acute for females during spring migration, since they must pay the oxidative costs of flight while preparing for reproduction; however, little previous work has examined how the antioxidant system of female spring migrants responds to dietary antioxidants and the oxidative challenges of regular flying. We fed two diets to female European starlings, one supplemented with a dietary antioxidant and one without, and then flew them daily in a windtunnel for two weeks during the fall and spring migration periods. We measured the activity of enzymatic antioxidants (GPx, SOD, CAT), non-enzymatic antioxidant capacity (ORAC), and markers of oxidative damage (protein carbonyls and lipid hydroperoxides) in four tissues: pectoralis, leg, liver, and heart. Dietary antioxidants affected enzymatic antioxidant activity and lipid damage in the heart, non-enzymatic antioxidant capacity in the pectoralis, and protein damage in leg muscle. In general, birds fed less antioxidants appear to incur increased oxidative damage while upregulating non-enzymatic and enzymatic antioxidant activity, though these effects were strongly tissue-specific. We also found trends for diet x training interactions for enzymatic antioxidant activity in the heart and leg. Flight-training may condition the antioxidant system of females to dynamically respond to oxidative challenges, and females during spring migration may shift antioxidant allocation to reduce oxidative damage.

Opis:
Mushrooms can be used in diet as nutraceutical or functional foods for promoting and maintaining health quality and health longevity. Mushrooms just like medicinal plants can be utilized in the form of powder or extract as curative agents of several diseases and in providing a healthy diet. In current study, ten different mushroom species were collected from Azad Jammu and Kashmir. The selected mushrooms were assessed for their nutritional value, biological activities and fatty acid profiling. Before extraction, proximate analysis of all selected mushrooms was performed. Protein, fat, ash and total carbohydrate contents ranged from 10-30%, 0.43-8.08%, 1.1-9.14%, 12.47-92.52%, respectively. Then mushrooms were extracted with three different solvents n-hexane, ethyl acetate and methanol for the assessment of their antioxidant potentials through different antioxidant assays like total phenolics, total flavonoid contents and DPPH inhibition assay. The order of antioxidant potential in different solvent is methanol>Ethyl acetate>n-hexane. Quantification of fatty acids by GC/MS analysis revealed that stearic acid, palmitic acid, oleic acid, linolenic acid were present in significant quantity in different mushrooms and unsaturated fatty acids were found abundant over saturated fatty acids in all the selected mushrooms. So presence of fatty acids in studied samples is one of the main biologically active ingredients from mushrooms. The study concluded that the selected mushrooms from different regions of Azad Jammu and Kashmir are of nutritional importance having significant antioxidant potential and contains several unsaturated fatty acids which contribute to the nutritional value of mushrooms.

Opis:
Flavonoids exhibit prooxidant cytotoxicity in mammalian cells due to the formation of free radicals and oxidation products possessing quinone or quinomethide structure. However, it is unclear how the cytotoxicity of flavonoids depends on the ease of their single-electron oxidation in aqueous medium, i.e., the redox potential of the phenoxyl radical/phenol couple. We verified the previously calculated redox potentials for several flavonoids according to their rates of reduction of cytochrome c and ferricyanide, and proposed experimentally-based values of redox potentials for myricetin, fisetin, morin, kaempferol, galangin, and naringenin. We found that the cytotoxicity of flavonoids (n=10) in bovine leukemia virus-transformed lamb kidney fibroblasts (line FLK) and murine hepatoma (line MH-22a) increases with a decrease in their redox potential of the phenoxyl radical/phenol couple and an increase in their lipophilicity. Their cytotoxicity was decreased by antioxidants and inhibitors of cytochromes P-450, α-naphthoflavone and isoniazide, and increased by an inhibitor of catechol-O-methyltransferase, 3,5-dinitrocatechol. It shows that although the prooxidant action of flavonoids may be the main factor in their cytotoxicity, the hydroxylation and oxidative demethylation by cytochromes P-450 and O-methylation by catechol-O-methyltransferase can significantly modulate the cytotoxicity of the parent compounds.

Opis:
There is a great diversity of the acute drugs overdose cases in clinical toxicology. Clinical situation is complicated by the coexistence of factors predisposing to the development o f adverse drug reactions (chronic use of drugs, polypharmacy, alcohol or drugs dependence, nutritional disorders) and by the presence of chronic organ damage, especially the liver and the kidney. The aim of this study w as to evaluate whether there are sensitive plasma markers belonging to the antioxidant system in patients exposed to various xenobiotics. We measured the activity of antioxidant parameters: catalase (CAT), glutathione peroxidase (GPX3), glutathione (GSH), sulfhydryl groups (-SH), carbonyl groups (=CO) and free radicals (2,2-diphenyl-1-picrylhydrazyl, DPPH, assay) in serum of 49 patients with acute intoxication caused by carbamazepine (CBZ, n = 9), mixed drug intoxication (MDI) (n = 9), alcohol withdrawal syndrome (AWS, n = 9), acetaminophen (APAP, n = 7), tricyclic antidepressants (TCAs) (n = 5), valproic acids (VA, n = 4), narcotics (N, n = 3), and three others (benzodiazepines, BZD, n = 2; barbiturates, n = 1). The results were compared w ith the parameters of not intoxicated patients (n = 39). All patients had lower catalase activity in comparison to the control group (41.9 ± 16.5 vs. 196.0 ± 82.2 U/mg protein, p = 0.000), while the increase of GSH level was particularly apparent only in patients with AWS (391.3 ± 257.9 pmol/mg protein) compared to the control group (171.4 ± 88.4 pmol/mg protein, p = 0.034) and to patients intoxicated with carbamazepine (152.8 ± 102.5 pmol/mg protein, p = 0.027). Some differences, but without statistical significance, were also observed in GPX3 activity between different groups of poisoned patients.
