Opis:
Novel series of 4-substituted 6,8-dibromo-2-(4-chloro-phenyl)-quinazoline have been designed and synthesized. All new derivatives were tested in vitro against MCF-7. Compounds Xc and XIb exerted powerful cytotoxic activity with low IC50 (6.3 and 6.9 µM) compared to doxorubicin 7.72 µM. Compounds Xa, IXb and IXc showed moderate cytotoxic effects with IC50 range (10.0 – 16.7) µM, respectively. Compounds IXa, XIc, XIa, VIIb, VIIIc, Xb and VIIIa showed promising cytotoxic effects with IC50 range (20.3 – 40 µM, respectively. ). Exploring their apoptotic effect; all compounds activated apoptotic cascade in MCF-7. Compounds Xc and XIb increased CASP3 activity more than doxorubicin.

Często opisywany jest również wpływ stanu zdrowia czy też aktywności społecznej na odczuwany przez jednostki poziom satysfakcji z życia. O wiele rzadziej spotykamy badania wskazujące na związek uczenia się ze zdrowiem, dobrym samopoczuciem czy satysfakcją z życia. Celem artykułu jest analiza wpływu uczenia się na odczuwaną satysfakcję z życia przez osoby w wieku 50 lat i więcej na podstawie wyników 8 rundy badania SHARE (The Survey of Health, Ageing and Retirement in Europe).
Scientific literature covers numerous studies carried out at showing the impact of learning on professional activity, earnings, position, etc. The effect of health or social activity on the level of life satisfaction is also one of the most often chosen aim of the research. Much less frequently we encounter studies indicating the relationship between learning and health, well-being or life satisfaction. The aim of the article is to analyze the impact of learning on the life satisfaction of people aged 50 and over based on the results of the 8th round of the SHARE (The Survey of Health, Aging and Retirement in Europe).

Opis:
Juglone is an allelopathin secreted by black walnut tree of the Juglandaceae family and is used as an active ingredient in many herbal preparations and as a commercial dye. It is considered as an important phytochemical with wide therapeutic potential. Black walnut extract has long been used in folk medicine to treat various types of cancers. It demonstrates antiviral, antibacterial, antifungal, and antitumor activities. The present study aimed to analyze the effect of juglone on the viability and proliferation of melanoma cells of C-32 (amelanotic melanoma) and COLO 829 (melanotic melanoma) cell lines in vitro and on the mRNA expression of genes encoding the proapoptotic BAX protein and caspase 3 and the gene encoding antiapoptotic BCL2 protein. The results showed a dosedependent effect of juglone on the viability, proliferation, and death induction in C-32 and COLO 829 melanoma cells and in HFF-1 normal dermal fibroblasts in in vitro cultures, but melanoma cells were more sensitive to juglone. Our findings revealed different mRNA expression patterns for all the studied genes in melanoma and normal cells treated with juglone in in vitro cultures.

Role of environmental specificity in CASP results

Background: Recently, significant progress has been made in the field of protein structure prediction by the application of artificial intelligence techniques, as shown by the results of the CASP13 and CASP14 (Critical Assessment of Structure Prediction) competition. However, the question of the mechanism behind the protein folding process itself remains unanswered. Correctly predicting the structure also does not solve the problem of, for example, amyloid proteins, where a polypeptide chain with an unaltered sequence adopts a different 3D structure. Results: This work was an attempt at explaining the structural variation by considering the contribution of the environment to protein structuring. The application of the fuzzy oil drop (FOD) model to assess the validity of the selected models provided in the CASP13, CASP14 and CASP15 projects reveals the need for an environmental factor to determine the 3D structure of proteins. Consideration of the external force field in the form of polar water (Fuzzy Oil Drop) and a version modified by the presence of the hydrophobic compounds, FOD-M (FOD-Modified) reveals that the protein folding process is environmentally dependent. An analysis of selected models from the CASP competitions indicates the need for structure prediction as dependent on the consideration of the protein folding environment. Conclusions: The conditions governed by the environment direct the protein folding process occurring in a certain environment. Therefore, the variation of the external force field should be taken into account in the models used in protein structure prediction.

Opis:
CASP (Critical Assessment of protein Structure Prediction) is a community-wide experiment for protein structure prediction taking place every two years since 1994. In CASP 11 held in 2014, according to the official CASP 11 assessment, our method named `nns' was ranked as the second best server method based on models ranked as first out of 81 targets. In `nns', we applied the powerful global optimization method of conformational space annealing to three stages of optimization, including multiple sequence-structure alignment, three-dimensional (3D) chain building, and side-chain remodeling. For the fold recognition, a new alignment method called CRF align was used. The good performance of the nns server method is attributed to the successful fold recognition carried out by combined methods including CRF align, and the current modeling formulation incorporating accurate structural aspects collected from multiple templates. In this article, we provide a successful example of `nns' predictions for T0776, for which all details of intermediate modeling data are provided.

Do all savings matter equally? : saving types and emotional well ‑being among older adults : evidence from panel data

Ill-being and mental ill-health have been on the rise in both Europe and the United States, especially among middle-aged and older adults. Although financial security has been shown to play a protective role in emotional well-being, little is known about the protective role of different types of family assets on mental health and well-being. Using longitudinal survey data from the Survey of Health, Aging and Retirement in Europe (SHARE) collected between 2004 and 2017, we examined the role of different types of family assets in emotional well-being and depression. A multivariate proportional hazard model with time-varying covariates was used. We found that family assets may play a significant protective role against depression, loneliness, and a decreased quality of life. Different forms of family assets may play diverse roles in protecting against the risks of ill-being and mental ill-health; however, their roles in increasing the chances of overcoming ill-being are less pronounced. Promotion of saving behaviours and proper financial management can help protect against adverse well-being and health outcomes in middle-aged and older adults.
