Assessment of semiquantitative parameters of dynamic contrast-enhanced perfusion MR imaging in differentiation of subtypes of renal cell carcinoma

BACKGROUND: To assess semiquantitative parameters of dynamic contrast-enhanced perfusion MR imaging (DCE) in differentiation of subtypes of renal cell carcinoma (RCC). MATERIAL AND METHODS: Prospective study conducted upon 34 patients (27 M, 7 F, aged 25-72 ys: mean 45 ys) with RCC. Abdominal dynamic contrast-enhanced gradient-recalled echo MR sequence after administration of gadopentetate dimeglumine was obtained. The time signal intensity curve (TIC) of the lesion was created with calculation of enhancement ratio (ER), and washout ratio (WR). RESULTS: The subtypes of RCC were as follows: clear cell carcinomas (n=23), papillary carcinomas (n=6), and chromophobe carcinomas (n=5). The mean ER of clear cell, papillary and chromophobe RCC were 188±49.7, 35±8.9, and 120±41.6 respectively. The mean WR of clear cell, papillary and chromophobe RCCs were 28.6±6.8, 47.6±5.7 and 42.7±10, respectively. There was a significant difference in ER (P=0.001) and WR (P=0.001) between clear cell RCC and other subtypes of RCC. The threshold values of ER and WR used for differentiating clear cell RCC from other subtypes of RCC were 142 and 38 with areas under the curve of 0.937 and 0.895, respectively. CONCLUSIONS: We concluded that ER and WR are semiquantitative perfusion parameters useful in differentiation of clear cell RCC from chromophobe and papillary RCCs.

In this study we have investigated the impact of differentiation of neuronal cells on their sensitivity to microbial toxins. We used the human neural crest-derived tumor cell line Paju, which can be induced to differentiation in vitro by treatment with phorbol 12-myristate 13-acetate. Addition of the highly toxic potassium ionophores cereulide (4.5 and 9.0 ng/ml) or valinomycin (20 ng/ml), to cultures of undifferentiated Paju cells caused collapse of the mitochondrial membrane potential - measured with the fluorescent probe 5,5',6,6'-tetrachloro-1,1',3,3'-tetrabenzimidazole carbocyanine iodide (JC-1) followed by detachment of the cells and their apoptotic death. After induced differentiation of the Paju cells, their mitochondria retained the membrane potential upon exposure to the toxins and the cells displayed increased resistance to apoptosis as compared with undifferentiated cells. This effect may be caused by an elevated expression of the anti-apoptotic protein Bcl-2 and of the neuroprotective factor, stanniocalcin, in differentiated cells.

Porcine ovarian cortex-derived putative stem cells can differentiate into endothelial cells in vitro

Opis:
Endothelial cells (ECs), the primary component of the vasculature, play a crucial role in neovascularization. However, the number of endogenous ECs is inadequate for both experimental purposes and clinical applications. Porcine ovarian putative stem cells (poPSCs), although not pluripotent, are characterized by great plasticity. Therefore, this study aimed to investigate whether poPSCs have the potential to differentiate into cells of endothelial lineage. poPSCs were immunomagnetically isolated from postnatal pig ovaries based on the presence of SSEA-4 protein. Expression of mesenchymal stem cells (MSCs) markers after pre-culture, both at the level of mRNA: ITGB1, THY, and ENG and corresponding protein: CD29, CD90, and CD105 were significantly higher compared to the control ovarian cortex cells. To differentiate poPSCs into ECs, inducing medium containing vascular endothelial growth factor (VEGF), basic fibroblast growth factor (bFGF), insulin-like growth factor (IGF), epidermal growth factor (EGF), ascorbic acid, and heparin was applied. After 14 days, poPSC differentiation into ECs was confirmed by immunofluorescence staining for vascular endothelial cadherin (VECad) and vascular endothelial growth factor receptor-2 (VEGFR-2). Semi-quantitative WB analysis of these proteins confirmed their high abundance. Additionally, qRT-PCR showed that mRNA expression of corresponding marker genes: CDH5, KDR was significantly higher compared with undifferentiated poPSCs. Finally, EC functional status was confirmed by the migration test that revealed that they were capable of positive chemotaxis, while tube formation assay demonstrated their ability to develop capillary networks. In conclusion, our results provided evidence that poPSCs may constitute the MSC population in the ovary and confirmed that they might be a potential source of ECs for tissue engineering.

Tumor Necrosis Factor-alpha (TNFα) is a pleiotrophic cytokine, affects either normal or tumor cells, and influences cellular differentiation. TNFα role in female reproduction has been proven to be mediated through an influence on prostaglandin (PGs) synthesis and output. To evaluate the possible role of TNFα in an auto-/paracrine regulation in the cat uterus, mRNA expression coding for TNFα and its receptors (TNFR1 and TNFR2), and TNFα protein content at different stages of the estrous cycle were investigated. Additionally, TNFα involvement in PG secretion at different stages of the estrous cycle was investigated by in vitro tissue culture. Gene expressions coding for TNFα and TNFR1 were the highest at diestrus (P < 0.05). TNFα protein expression was the lowest at interestrus (P < 0.05). Nevertheless, TNFR2 was not affected by the estrous stage. TNFα at a dose of 1 ng/ml significantly increased PGF₂α secretion at estrus (P < 0.01) and PGE₂ secretion at diestrus (P < 0.001) after 12h incubation. Overall findings indicate that TNFα locally produced in the cat’s uterus, stimulates PG secretion in an estrous cycle-related manner.

Influence of calcium ions and confluence of squamous cell carcinoma cell line A431 on transcript and protein level of MCPIP1
Wpływ jonów wapnia oraz konfluencji komórek płaskonabłonkowego raka skóry linii A431 na poziom transkryptu i białka MCPIP1

Głównym regulatorem różnicowania komórek naskórka jest wapń. Jony wapnia wpływają na obniżenie potencjału proliferacyjnego, zahamowanie migracji komórek oraz przyspieszenie procesów różnicowania. Białko MCPIP1 bierze udział podczas różnicowania wielu typów komórek, w tym keratynocytów pierwotnych oraz komórek linii HaCaT. Komórki płaskonabłonkowego raka naskórka inkubowano z jonami wapnia o stężeniu 1,8 mM przez 24h, 48h i 72h, a następnie sprawdzano zmiany MCPIP1 na poziomie białka metodą Western blot i na poziomie transkryptu metodą qRT-PCR. Podczas długich inkubacji komórek zwiększa się ich konfluencja, dlatego też badano sam proces wzrostu konfluencji na zmiany poziomu białka MCPIP1. Uzyskane wyniki wskazują, że poziom białka MCPIP1 (formy 66 kDa i 36 kDa) spada natomiast poziom transkryptu rośnie wraz z wydłużającym się czasem inkubacji z jonami wapnia,. Przy wzrastającej konfluencji komórek, poziom białka MCPIP1 (66 kDa) zwiększa się, natomiast poziom mniejszej formy białka (36 kDa) maleje.
The main regulator of epidermal cell differentiation is calcium. Calcium ions reduce proliferative capacity, inhibit cell migration, and accelerate differentiation processes. The MCPIP1 protein is involved in the differentiation of many cell types, including primary keratinocytes and HaCaT cells.Squamous cell carcinoma cells were incubated with 1.8mM calcium ions for 24h, 48h, and 72h, and then MCPIP1 changes were monitored at the protein level by the Western blot method and at the transcript level by the qRT-PCR method. During long incubation of cells, their confluence is increased, therefore the confluence growth process was investigated for changes in MCPIP1 protein level. The results showed that the level of MCPIP1 (66 kDa and 36 kDa forms) decrease while the level of transcript increases with the longer incubation time with calcium ions. During the increasing cell confluence, the level of the MCPIP1 protein (66 kDa) increases while the level of the smaller protein form (36 kDa) decreases.
