Opis:: This paper discusses current interpretative differences between such terms as: coupled, complex, multidimensional, and multiple disabilities. The analyses of these terms and what they connote are also presented in the light of the concepts of human disability proposed by the World Health Organization from 1980 and 2001. Moreover, based on the presented analyses, the possible interpretations of the selected terms are discussed in relation to the required educational and rehabilitative needs of individuals with particular types of disabilities.

Opis:: The aim of this article is to investigate the influence of intersectional politics of recent disability drama on the conceptualization of place. My point of departure is Una Chaudhuri’s concept of geopathic place and its discussion within the context of dramaturgy of disability by Victoria Ann Lewis. In her Staging Place: The Geography of Modern Drama, Una Chaudhuri claims that ‘the geopathic paradigm underlying realist drama … supports a certain construction of identity: identity as a negotiation with – and on occasion a heroic overcoming of – the power of place’ (1997, p. 56). Victoria Ann Lewis in her 2004 article argues that artists with disabilities often ‘re-code’ identity and place as well as ‘disrupt the liberal dichotomies of individual freedom vs. confinement/prison and the related hierarchical oppositions of movement over stasis, and time over place.’ In my discussion of All of Us by Francesca Martinez, I will focus on very recent redefinitions of these oppositions and dichotomies enabled by intersectional constructions of characters and spaces. I will also trace how in exploring the relation between these two categories and human embodiment, All of Us promotes the notions of human variety and shared humanity, which prevent potential stigmatizations of characters with disabilities.

Opis:: A child with pervasive developmental disorder shows the disturbances in the developmental dynamics, an effect that might be strengthened if autism or intellectual disorders have been diagnosed as well. This article is an attempt to compare the condition of the perceptual functions of an eight-year-old boy who suffers from multiple developmental disorders, that is, autism and moderate intellectual disorder, to that of the developmental age of a six-year-old child. The empirical data contain numerous observations, the studies based on the self-prepared questionnaires assessing the child’s psychomotor development, and the interviews with parents, teachers, and therapists. The outcomes show the disproportion between the examined cases and indicate significant development delay of the child with autism – absent in his healthy friend – which results in the decreased participation in his environment.

Opis:: Niniejszy artykuł stanowi próbę zastosowania teorii interakcji do analizy zachowań 8-letniego dziecka ze sprzężonymi zaburzeniami rozwoju – autyzmem i niepełnosprawnością intelektualną w stopniu umiarkowanym. Autyzm, rozumiany jako całościowe zaburzenie rozwojowe, w zdecydowanej większości przypadków współwystępuje z niepełnosprawnością intelektualną, która dotyczy nie tylko sfery poznawczej, ale również możliwości społecznych i adaptacyjnych. Stąd w artykule podjęto próbę zróżnicowania mechanizmów doprowadzających do interferencji objawów zaburzeń interakcji dziecka ze sprzężonymi zaburzeniami rozwoju.
The article aims to implement the interaction theory in order to analyse the behaviour of an eight-year-old boy with multiple-complex developmental disorders – autism and moderate intellectual disability. Autism, also called pervasive developmental disorder, in most cases co-occurs with intellectual disability that refers not only to the cognitive sphere but also to social and adaptive opportunities. Thus, this study is an attempt to differentiate the mechanisms that lead to interference of the symptoms of the child’s interactions conditioned by multiple-complex developmental disorder.

Opis:: Cząsteczki tRNA są jednym z podstawowych elementów mechanizmu translacji białka. Aby regulować rygorystyczny proces translacji, konieczne są modyfikacje tRNA, aby utrzymać jego strukturę drugorzędową i, co ważniejsze, przyczynić się do parowania zasad między tRNA i mRNA w rybosomie. Jedna z najbardziej istotnych modyfikacji tRNA wpływa na pozycję wobble U_34 w pętli antykodonowej. Kompleks białkowy Elongatora (Elp) katalizuje modyfikację chemiczną na U_34 , aby wygenerować 〖cm〗^5 U_34 , które może być dalej modyfikowane do różnych produktów końcowych. Stwierdzono, że rozregulowanie Elp, takie jak nadekspresja lub utrata funkcji, wiąże się z początkiem nowotworów lub chorób neurodegeneracyjnych. Elp jest wysoce konserwatywny od organizmów drożdżowych po organizmy ludzkie i składa się z dwóch kompleksów, mianowicie Elp123 i Elp456. Elp123 zawiera aktywność katalityczną modyfikacji, natomiast Elp456 niesie aktywność ATPazy. Strukturę Elp i charakterystykę biochemiczną przedstawiono niedawno, ale badania te przeprowadzono dla Elp drożdżowego, natomiast struktury krystaliczne podjednostki Elp 3 uzyskano z bakterii i archeonów. Biochemiczna charakterystyka ludzkiego Elp nie została jeszcze wyjaśniona. W tej pracy magisterskiej zrobiono pierwszy krok w badaniu ludzkiego Elp456 w odniesieniu do profili ekspresji jego izoform w różnych ludzkich liniach komórkowych. Wyniki wykazały, że wzory ekspresji izoform nie były jednakowe w różnych typach komórek. Ponadto wybrano jeden spośród hElp4, hElp5 i hElp6 do ekspresji rekombinowanego białka w bakteryjnym lub owadzim systemie ekspresyjnym. Obecnie hElp4v1 został z powodzeniem sklonowany do wektora ekspresyjnego E. coli.
tRNA is one essential element of the protein translation mechanism. To regulate protein translation process, modifications of tRNAs are required to maintain its secondary structure and, more importantly, contribute to the base pairing between tRNA and mRNA in the ribosome. One of the most crucial tRNA modification affects the wobble position 〖( U〗_34) at the anticodon stem-loop. The Elongator protein complex (Elp) catalyzes the chemical modification on 〖(U〗_34), to generate the cm^5 U_34, which can be further modified to different end products. The dysregulation of Elp, such as overexpression or loss of function, have found to be related to the onset of cancers or neurodegenerative diseases. Elp is highly conserved from yeast to human and consists of two sub-complexes, namely Elp123 and Elp456. Elp123 contains the catalytic modification activity while Elp456 harbors ATPase activity. The Elp structure and biochemical characterizations were presented recently. Still, all were performed in the yeast system, whereas crystal structures of the Elp subunit three were obtained from bacteria and archaea. The biochemical characterization of human Elp is yet to be clarified. In this master thesis, we took the first step in investigating human Elp456 regarding their isoform expression profiles in different human cell lines. The results revealed that isoform expression patterns were not universal in different cell types. Furthermore, we have chosen variant one from hElp4, hElp5, and hElp6 for recombinant protein expression in either the bacterial expression system or insect cell expression system. Currently, hElp4v1 has been successfully cloned into the E. coli expression vector.
