The aim of the dissertation was to determine (select) the best growth medium for a basket willow (Salix viminalis L.) which contains optimal concentration of kinetin. The plant material taken from three varieties of willows: Bjor, Jorr and Tora, which grew in the experimental field, was sterilized and placed on a complete MS (Murashige and Skoog, 1962) growth medium. The propagated plants were used in an experiment, where MS substrate was enriched by growth regulator – kinetin, in various concentrations (0.5 mg∙dm-3, 1 mg∙dm-3, 2 mg∙dm-3 and 3 mg∙dm-3). The substrate which contained 0.5 KIN was considered to be the best for willow’s propagation, because plants cultivated with usage of this medium have shown the largest mass, the highest number of shoots and leaves, and also the highest quantitative and elongation growth of the roots. Among examined varieties of Salix, the best one was Bjor variety, where evaluated parameters were distinctive in comparison to two remaining varieties.

Opis:
The genus Petunia is a plant of high economic importance in the world-wide horticulture. These ornamental plants are often exposed to soil salinity that negatively affects their development. Chitosan is a biopolymer with multiple applications in plant breeding but it also minimizes the adverse effects of abiotic stresses on plant growth. The objective of this study was to investigate the effects of chitosan on petunia shoots development under salt stress in vitro. In the first experiment, four types of chitosan with molecular weight of 3.33, 8, 10 and 970 kDa in the concentrations of 0, 10, 15 and 20 ppm were supplemented into MS medium. In the second experiment, petunia shoots were grown on MS medium with the addition of different molecular weight of chitosan in the concentration of 15 ppm each and 100 mM NaCl. The results indicated that all of chitosan types and concentrations stimulate the plant growth in comparison to control. However, 15 ppm chitosan concentration was more effective than other concentrations used. Salinity caused a significant reduction in shoot and root length, fresh and dry mass, plant water contents, while chitosan (970 kDa) adjusted the salt toxicity. It is concluded that chitosan would be able to stimulate the growth of petunia shoots in vitro independent of their molecular weight. It was observed that the addition of chitosan of 970 kDa to MS medium under salinity conditions may alleviate the inhibitory effect of salt stress on the plant growth.

W ostatnich latach obserwuje się coraz powszechniej występujące zjawisko niszczenia lub samoistnego zamierania pomnikowych dębów szypułkowych (Quercus robur L.), których wiek sięga kilkuset lat. W takiej sytuacji kluczowe jest zachowanie zasobów genowych, szczególnie w przypadku sędziwych drzew, które obok znaczenia kulturowego mają wiele cennych cech odporności na zmieniające się czynniki środowiskowe, ukształtowane na przestrzeni setek lat. Metoda wegetatywnego rozmnażania dębów tradycyjnymi technikami nie jest możliwa, ponieważ pędy dębów nie ukorzeniają się poprzez zrzezy czy odkłady, a szczepienie nie daje jednorodnej genetycznie rośliny z pędem i korzeniem. Dodatkowo proces ukorzeniania się pędów jest szczególnie nieskuteczny w przypadku wiekowych dębów, dlatego niezbędne było sięgnięcie po techniki kultur tkankowych. W naszych badaniach testowaliśmy możliwość klonowania z wykorzystaniem metody in vitro 21 dębów pomnikowych z terenu Polski, w wieku 300–800 lat. Metoda ta polegała na pobraniu zdrewniałych pędów z pąkami śpiącymi, które następnie trzymano w hodowli wazonowej w fitotronie w celu uzyskania z nich pędów odroślowych. Fragmenty tych pędów odkażono i namnożono na pożywce agarowej, zawierającej niezbędne składniki odżywcze oraz regulatory wzrostu. Wybrane z hodowli in vitro pędy ukorzeniono na pożywce z dodatkiem węgla aktywnego. Tylko kilka z badanych dębów pomnikowych wykazywało potencjał do mikrorozmnażania. Były wśród nich i te najstarsze z testowanych drzew. Ukorzenione pędy po zahartowaniu przeniesiono w podłoże stałe (Chmielarz et al., 2016). W ten sposób sklonowano około 800–letni dąb Rus z Rogalina i Dąb Wybickiego z Muzeum Hymnu Narodowego w Będominie, liczący około 400 lat. W kwietniu 2019 r. otrzymane sadzonki, o wysokości około 2 m, posadzono w niedalekiej odległości od drzew matecznych.
In recent years a decline of the oldest oaks (often monumental trees) aged up to several hundred years has been observed in Poland. That is why conservation of their genetic resources is crucial. The trees have many valuable traits of resistance to changing environmental factors, which allowed them to survive in a changing environment. Vegetative propagation of oaks by using traditional methods is not possible because they cannot be easily propagated by cuttings. The main goal of our research was to assess the potential of the oldest oaks growing in Poland for regeneration in plant tissue culture. Plant material was collected from 21 monumental oaks. Their lignified shoots were collected at the end of April and next adventitious shoots were induced from them in vase culture. The shoots, used as explants in tissue culture, were propagated and rooted. As a result of this study, correctly growing plantlets (clones) were obtained under ex vitro conditions of several monumental oaks. Among them Rus Oak and The Wybicki Oak. There were no significant correlations between the age or trunk circumference at breast height of the maternal trees and the ability of their adventitious shoots to root in vitro culture, and the success rate of their acclimation. Two cloned monumental trees, Rus Oak and The Wybicki Oak were planted as 2 m high seedlings respectively in Rogalin and Będomin, near their mother trees in April, 2019.

A field experiment was carried out in two vegetation seasons in the years 2012 and 2013 on very light soil at Kruszyn Krajeński, in the vicinity of Bydgoszcz, Poland. The aim of the study was to characterize morphological achenes and determine the fertility potential and real of cup plant (Silphium perfoliatum L.) plants derived from micropropagation grown under irrigation. Low fertility real of cap plant resulted from the participation of infertile fruit-achenes it means devoid of the seeds. Due to the fact that this plant is entered on the list of invasive species, this property may be desirable during plantation establishment. As a result, the plant will not be so easy to penetrate into the ecosystem and will not pose a threat to native biodiversity.

Opis:
A vanishing species in Poland - Plantago maritima L. was regenerated in vitro from tips of shoots (obtained in vitro) and from different explants of 4-week-old seedlings: seedling tips, hypocotyls, cotyledons, roots. Murashige and Skoog basal medium, supplemented with 0.6 pM indole-3-acetic acid in combination with cytokinins 6-benzyladenine, zeatin or kinetin, was used. The plants obtained in the result of micropropagation were normal in appearence. It was proved that Plantago maritima species was amenable to propagation from different kinds of explants. The method may be of significance for protection of sea plantain.

The micromorphological studies of in vitro and field transferred plants of Duranta erecta were conducted in the present investigation. The nodal shoot segments were used as explants for the establishment of cultures. The shoots were multiplied on MS medium (Murashige and Skoog) supplemented with 6-benzylaminopurine (BAP), kinetin (Kin) and Naphthalene acetic acid (NAA). In vitro multiplied shoots were rooted on half strength MS medium augmented with indole-3 butyric acid (IBA). The rooted plantlets were successfully hardened and field transferred. A comparative foliar micromorphological studies were made of in vitro grown and field transferred plants to understand the developmental changes of micro-external structures of plants towards environment. The differences in leaf constants such as stomata, vein-islets and termination numbers and trichomes across the environments imply the developmental changes of micropropagated plants of Duranta erecta.

The aim of the study was to determine an optimal composition of growth media for Drosera rotundifolia L. cultured in vitro. Adventitious shoots of Drosera rotundifolia originating from an established in vitro culture were placed on complete Murashige and Skoog (MS) (1962) medium, MS medium with nutrient content reduced to ¾ MS, ½ MS, ¼ MS, 1/6 MS and MS medium with selected macronutrient (KNO3, KNO3 + NH4NO3, KH2PO4 or CaCl2) content reduced by ½. The medium containing ¼ of standard MS nutrients was found the most effective, as plants in this variant had the greatest weight and developed the greatest number of adventitious shoots (37.1). It was also conducive to root growth and development. Flowering was only observed on complete MS, ¾ MS and ½ MS media and MS with reduced content of KNO3 or KNO3 + NH4NO3. Reducing mineral content to ¼ MS or more resulted in red coloration of glandular tentacles.

The plant material were plants of sandy everlasting (Helichrysum arenarium (L.) Moench) collected from natural locations in October 2012. Initial explants were apical buds enfolded into two leaves. Isolated explants were chemically sterilized. Reproducted shoots were divided and put into three types of mediums: medium MS (control) containing 37 mg P·dm-3, medium MS without additive of P (0,0 mg P·dm-3), medium MS with additional amound of P (74 mg P·dm-3). The aim of research was to specify the influence of phosphorus content in medium for choosen growth parameters and accumulation of assimilatory pigments: chlorophyll a, b, chlorophyll a+b, carotenoids. Analysis of these compounds were done spectrophotometrically. The made research, proved, that the richest in mentioned substances were microseedlings growing on the medium with increased amount of phosphorus. Phosphorus deficiency in medium MS had a statistically essential effect on changes in growth modifications of microseedlings of sandy everlasting (Helichrysum arenarium (L.) Moench). Additional amount of phosphorus in the medium had a statistically essential effect on increasing of chlorophyll a+b in comparison with plants growing on the medium MS with optimal phosphorus amount or without this element by accordinglly 36% and 23%. Moreover, it caused a growth of content of chlorophyll a, b and carotenoids by accordinglly 18%, 32%, 20% in comparison with variant, where the medium MS without phosphorus was applied.

Human activity has caused environmental transformations for millennia, most often driven by the desire to acquire natural resources. Unfortunately, the desire to acquire these resources in many cases results in a reduction in biodiversity, that is, a reduction in genetic diversity in a given area. In addition, the transformation of the environment results in the replacement of plant species that originally existed in a given area by other vegetation, including foreign ones. Environmental transformation can take various forms, such as agricultural intensification, the acquisition of new mining areas, or simply the occupation of more land for municipal or industrial purposes. Nor can we forget one of the most important problems of recent decades: global warming, which contributes significantly to climate change and leads to the disappearance of many plant species. The way to reduce the negative human impact is to carry out environmental protection activities, both passive and active. In this paper, we take a closer look at how in vitro cultures of plants can benefit biodiversity conservation thanks to the possibility of micropropagation and long-term storage of plants.
