Opis:
Wielkopąkowiec porzeczkowy (Cecidophyopsis ribis), jest jednym z najgroźniejszych szkodników porzeczki czarnej. Powoduje on deformację pąków porzeczki czarnej i redukuje plon owoców. Wybranie najkorzystniejszych markerów molekularnych sprzężonych z odpornością na wielkopąkowca porzeczkowego może przyspieszyć proces hodowlany i poprawić skuteczność przy wyborze genotypów odpornych na tę chorobę. Celem pracy była analiza regionów genomu sprzężonych z odpornością porzeczki czarnej (Ribes nigrum) na wielkopąkowca porzeczkowego poprzez ocenę stopnia polimorfizmu w grupach sprzężeń obejmujących regiony Ce i P i poszukiwanie nowych fragmentów genomu regulujących odporność na C. ribis. W wyniku analizy elektroforegramów z produktami reakcji CAPS, zidentyfikowano 119 polimorficznych fragmentów DNA, z których wytypowano 100 do analizy sekwencji. W wyniku sekwencjonowania uzyskano 9 specyficznych odczytów na matrycach DNA odmian ‘Ceres’, ‘Ojebyn’, ‘Vir’, ‘Ores’, ‘Foxendown’, ‘Ben Finlay’ i ‘Bona’.
Blackcurrant gall mite (Cecidophyopsis ribis) is one of the most dangerous pests of blackcurrant. It causes deformation of blackcurrant buds and reduces fruit yield. The selection of the most favorable molecular markers coupled to resistance to C.ribis, can accelerate the breeding process and improve efficacy when choosing genotypes that are resistant to this disease. The aim of this study was to analyze genome regions conjugated with resistance of blackcurrant (Ribes nigrum) to blackcurrant gall mite by assessing the degree of polymorphism in coupling groups, including the Ce and P regions, and by searching for new genome fragments regulating C. ribis resistance. As a result of the analysis of electrophoregrams with CAPS reaction products, 119 polymorphic DNA fragments were identified, out of which 100 were selected for sequence analysis. As the end product of sequencing, 9 specific readings were obtained on DNA matrices of the varieties ‘Ceres’, ‘Ojebyn’, ‘Vir’, ‘Ores’, ‘Foxendown’, ‘Ben Finlay’ and ‘Bona’.

Opis:
Sweet cherry is a vegetatively propagated, perennial plant with high level of heterozygosity and ancient breeding history. Therefore, it is necessary to keep, conserve and evaluate known genetic resources for future breeding programs and fruit production stability. In present, the utilization of DNA molecular genetic analyses is the best suitable method for evaluation of individual accessions, thus we eliminated duplications and characterized the genetic relationships. In our work, we used PCR primer combinations for 19 SSR and 2 EST-SSR loci for analyses of 123 current, old and local sweet cherry cultivars from Czech genetic resources of Research and Breeding Institute of Pomology in Holovousy. In total, 115 polymorphic fragments were amplified, which we used for hierarchical cluster analysis of genetic variability. The result dendrograms were divided into three main clusters and ten subgroups. Clustering corresponded to genealogical and geobotanical characteristics of individual accessions as breeding history of several known accessions.

A total of 96 indigenous Brassica rapa accessions were collected from different locations of Khyber Pakhtunkhwa, Pakistan. Simple Sequence Repeats (SSR) markers were used to identify the most diverse genotypes among the collected lots. Twenty six (26) different SSR primers were used for (genetic) variability among collected genotypes. These primers were selected from literature based on their previous results. These primers produced 135 scorable bands of which 75 were polymorphic, with an average of 55.5% polymorphic loci, and reflected the broader genetic background of the collected genotypes. An average 2.88 polymorphic bands with an average PIC value of 0.49 was recorded. Unweighted Pair Group Method with Arithmetic Mean (UPGMA) divided all genotypes into three main groups. Group one contained three clusters, while group two and three had four and two clusters each. Based on the UPGMA dendrogram, genotypes collected from Kohat, Bannu, Swat and Haripur showed considerable amount of variation. From the present study, it is concluded that SSR markers can be proved as the best tool for the genetic variability of other local and exotic B. rapa genotypes.

Opis:
Hybridization of a multi-locus DNA probe, R 18.1, to genomic DNA from poultry showed a highly polymorphic fingerprint pattern. The detected DNA fingerprints are individually specific and differ between Green Legged Patridgenous (GLP) and Rhode Island Red (RIR) stock of chickens. The average numbers of detected bands in RIR were 18.68 and in GLP - 15.33, but the average band sharing levels were 0.619 and 0.431, respectively. The level of polymorphism may be connected possibly with a higher level of inbreeding in the examined stock of chicken.

Provenance trials were designed to analyse the quantitative responses of tree species to environmental variables found in different experiment location. However, we have still limited knowledge how natural and artificial selection affects genetic variation of the species populations gather in such experimental sites. We have used bulked DNA-based RAPD and ISJ analysis to investigate genetic diversity and differentiation of Scots pine populations from two Polish locations of IUFRO 1982 provenance trial placed in Kórnik and in Supraśl. Applied categories of DNA markers differed in terms of revealing genetic diversity of the species. Ten RAPD primers applied in the study yielded a total of 75 bands, of which 21 (28%) and 15 (20%) were polymorphic in Kórnik and in Supraśl, respectively. Six ISJ primers revealed 42 bands of which 4 (9.52%) and 14 (33.3%) were polymorphic in Kórnik and in Supraśl, respectively. The genetic diversity and differentiation was low, as expressed by He=0.071 and He=0.085, and by genetic distance values which ranged from 0.0 to 0.240 (on average 0.081) and from 0.017 to 0.188 (on average 0.094) for Kórnik and Supraśl, respectively. Location of provenance trial appeared to have a significant influence on revealed level of genetic polymorphism and pattern of interpopulation differentiation. However, genetic structure found for analysed Scots pine provenances from IUFRO 1982 in Kórnik was also confirmed for Supraśl experimental site. In the light of available data we also discussed the influence of historical migration routes and gene flow on observed genetic variation of the species.

Bread wheat is a major food crop on a global scale. Stripe rust, caused by Puccinia strii- formis f. sp. tritici, has become one of the largest biotic stresses and limitations for wheat production in the 21st century. Post 2000 races of the pathogen are more virulent and able to overcome the defense of previously resistant cultivars. Despite the availability of effective fungicides, genetic resistance is the most economical, effective, and environmen- tally friendly way to control the disease. There are two major types of resistance to stripe rust: all-stage seedling resistance (ASR) and adult-plant resistance (APR). Although both resistance types have negative and positive attributes, ASR generally is race-specific and frequently is defeated by new races, while APR has been shown to be race non-specific and durable over time. Finding genes with high levels of APR has been a major goal for wheat improvement over the past few decades. Recent advancements in molecular mapping and sequencing technologies provide a valuable framework for the discovery and validation of new sources of resistance. Here we report the discovery of a precise molecular marker for a highly durable type of APR – high-temperature adult-plant (HTAP) resistance locus in the wheat cultivar Louise. Using a Louise × Penawawa mapping population, coupled with data from survey sequences of the wheat genome, linkage mapping, and synteny analysis techniques, we developed an amplified polymorphic sequence (CAPS) marker LPHTAP2B on the short arm of wheat chromosome 2B, which cosegregates with the resistant pheno- type. LPHTAP2B accounted for 62 and 58% of phenotypic variance of disease severity and infection type data, respectively. Although cloning of the LPHTAP2B region is needed to further understand its role in durable resistance, this marker will greatly facilitate incorpo- ration of the HTAP gene into new wheat cultivars with durable resistance to stripe rust.

Drosera L. is a genus of carnivorous plants that comprises approximately 250 species, although this number is probably not complete. Some of these taxa exhibit only small differences in morphological traits that can be partly influenced if the taxa are propagated in vitro. Here, we focus on the verification of putative clones of Drosera spathulata Labill., Drosera rotundifolia L. and Drosera binata var. Dichotoma species cultivated in vitro using molecular markers covering the internal transcribed spacer (ITS) region of 45S ribosomal DNA (rDNA). Following the polymerase chain reaction (PCR) amplification of ~360-bp DNA fragments and sequencing, the sequences were aligned with corresponding sequences in the National Center for Biotechnology Information (NCBI) database. In addition, each of tested PCR amplicons had a specific restriction profile that predominantly enables the differentiation of D. rotundifolia and D. spathulata; the shape of the leaves does not have to be a clear morphologically distinguishable trait.
