Opis:
Although Hedera helix is the only native ivy distributed in Central Europe, other ivy species are cultivated in this region and have horticultural importance, including H. hibernica, H. colchica, and H. azorica. On the basis of morphological, cytological, and phenological studies, a newly identified diploid species of ivy, H. crebrescens Bényei-Himmer & Höhn, was recently described from Hungary. Due to its high reproductive capacity and vigorous growth, this species can be considered a potentially invasive plant that could readily supplant H. helix in its native habitats. To characterize the molecular taxonomic status of H. crebrescens, we conducted a molecular genetic analysis based on five chloroplast and one nuclear DNA regions. Our phylogenetic reconstruction supported the monophyly of Hedera, with a tree topology similar to that previously obtained based on phylogenetic cpDNA analyses. Mediterranean species of ivy were well separated from the remainder of the European species, as well as from Asian species. Hedera crebrescens represented a single independent haplotype within the Asian–European cluster, whereas H. helix proved to be polyphyletic. The detected species-specific haplotype and invariability among studied specimens obtained from different geographical locations, provide support for the taxonomical autonomy of H. crebrescens.

Genetic variability of three sympatric crucian carp ( Carassius carassius ) populations from NW Poland was studied within a research project aimed at assessing the utility of those populations for stocking in inland waters. DNA samples were collected from 65 individuals. Restriction analysis was performed using 4 enzymes (HaeIII, HinfI, FspBI, TasI) of known restriction sites. The restriction pro files obtained were classified as belonging to a single haplotype (H-1). Selected DNA products were sequenced; the subsequent comparison made it possible to detect the presence of substi- tutions in the genome fragment analysed.

This study reports the propagation dynamics of the Kara Sea surface desalinated layer (SDL) during the summer and autumn seasons. We analysed shipboard measurements data collected in 2013–2018 and MODIS ocean colour data that correlated with the shipboard ones. We formulated a comparatively strict criterion to determine the SDL border based on satellite data. For that, we analyzed the shipboard flow-through measuring system data obtained while crossing the surface desalinated layer border. Further, we used a regional algorithm to process the satellite data and estimate the coloured dissolved organic matter absorption coefficient for the Kara Sea. The results demonstrate a significant effect of the wind regime on the interseasonal and interannual variability of the transformation of the SDL boundaries. The positions of the surface desalinated layer boundaries at different times during 2013–2018 are given. The obtained results are important for calculating the heat balance and analyzing the Kara Sea bio-productivity.

W pracy przedstawiono przegląd metod analizy ekspresji genów z zastosowaniem technik: mikromacierzy DNA, seryjnej analizy ekspresji genów (SAGE), jak również odwrotnej transkrypcji – RT-PCR oraz qRT-PCR. Zaprezentowano możliwości wykorzystania poszczególnych technik wraz z przykładami ich praktycznego zastosowania w badaniach molekularnych rzepaku.
This paper presents an overview of gene expression analysis methods, including: DNA microarrays, SAGE (Serial Analysis of Gene Expression), as well as reverse transcription techniques: RT-PCR and qRT-PCR. It also includes some examples of application of the methods in molecular studies on oilseed rape genome and transcriptome.

Tiagabine is an antiepileptic drug used for the treatment of partial seizures in humans. Recently, this drug has been found useful in several non-epileptic conditions, including anxiety, chronic pain and sleep disorders. Since tachycardia—an impairment of cardiac rhythm due to cardiac ion channel dysfunction—is one of the most commonly reported non-neurological adverse effects of this drug, in the present paper we have undertaken pharmacological and numerical studies to assess a potential cardiovascular risk associated with the use of tiagabine. A chemical interaction of tiagabine with a model of human voltage-gated ion channels (VGICs) is described using the molecular docking method. The obtained in silico results imply that the adverse effects reported so far in the clinical cardiological of tiagabine could not be directly attributed to its interactions with VGICs. This is also confirmed by the results from the isolated organ studies (i.e., calcium entry blocking properties test) and in vivo (electrocardiogram study) assays of the present research. It was found that tachycardia and other tiagabine-induced cardiac complications are not due to a direct effect of this drug on ventricular depolarization and repolarization.

Till date, it is well-known that synthetic pyrethroids are safe to mammal but toxic to non-mammals. The present objective was an in silico study to detect oral acute toxicity, organ toxicity, immunotoxicity, genetic toxicity endpoints, nuclear receptor signalling, and stress response pathways of common synthetic pyrethroids by using ProTox-II webserver. The chemical compounds especially different synthetic pyrethroids such as pyrethrin I, Cinerin I and Jasmolin I (esters of Chrysanthemic acid), Pyrethrin-II, Cinerin II and Jasmolin II (Esters of Pyrethric acid), type I pyrethroids (esters without alpha-cyano group) such as allethrin, resmethrin, permethrin and bifenthrin and type II pyrethroids (esters with alpha-cyano group) such as fenvalerate, cyhalothrin, cypermethrin and deltamethrinwere selected from available literature. ProTox-II webserver was used for toxicological assessment in organism, organs, cell and gene level along with molecular mechanisms of toxicity. The predictive results for the toxicity of common synthetic pyrethroids compounds, Deltamethrin showed highly toxic compound among 14 compounds as fatal if swallowed as class II followed by Cypermethrin, Cyhalothrin, Bifenthrin, Resmethrin, Fenvalerate and Permethrin but hepatotoxic potential was only Deltamethrin and Fenvalerate while immunotoxic was obtained Permethrin. On the other hand, none of the compounds were obtained cytotoxic and carcinogenic but 9 compounds viz. Pyrethrin I, II Cinerin I, II, Jasmolin I, II, Allethrin, Resmethrin and Permethrin were observed mutagenic active. In case of NR signalling pathways, all compounds were inactive but eight compounds such as Pyrethrin I, II, Cinerin I, II, Jasmolin I, II, Allethrin and Resmethrin were obtained nrf2/ARE and HSE active while MMP active compounds were obtained Fenvalerate, Cyhalothrin and Deltamethrin respectively. For p53 and ATAD5 parameters, all fourteen compounds such as were obtained inactive. In conclusion, the present predictive results are suitable for academician, researchers, industries, etc. those who are making drugs and environmental chemicals. This web server helps faster screening of large numbers of compounds within short duration and no animal testing. This present in silico study easily detects toxin(s), which can be validated in future through in vitro and in vivo experimental assay.

While a healthy endothelium serves to maintain vascular haemostasis, a malfunctioning endothelium leads to various cardiovascular diseases, including atherothrombosis. Endothelial dysfunction is characterized by increased vascular permeability, impaired endothelium-dependent responses and various pro-inflammatory and pro-thrombotic changes in endothelial phenotype, all of which could provide the basis for an in vivo diagnosis of endothelial dysfunction. In the present review, we briefly summarize the magnetic resonance imaging (MRI)-based methods available for assessing endothelial function in animal models, especially in mice. These methods are aimed to assess biochemical phenotype using molecular imaging, endothelium-dependent responses or changes in endothelial permeability. All these approaches provide a complementary insight into the endothelial dysfunction in vivo and may offer a unique opportunity to study endothelium-based mechanisms of diseases and endothelial response to treatment.

Anti-atherosclerotic activity of catechins depends on their stereoisomerism

In terms of stereochemistry, catechins are divided into two groups: (-) epi forms (2R, 3R) and (-) forms (2S, 3R). Most of the catechins present in green tea are (-) epi forms (2R, 3R). Under the influence of high temperatures, in anaerobic conditions, as a result of epimerization the proportion of the (-) form (2S, 3R) increases. The data indicate that the presence of thermally modified catechins in the diet more efficiently reduces the development of atherosclerosis in apoE knockout mice than the presence of native catechins. The addition of the thermally modified formulations to the high-fat diet resulted in a reduction of the area of atherosclerotic lesions by about 28% (en face method) and 45% (cross-section method) compared to the group fed the high-fat diet without catechins. Furthermore, the body weight gain and plasma TBARS concentration in mice fed a diet with the addition of catechins depends on the degree of epimerization of catechins and decreases with increasing content of catechins belonging to the (-) form (2S, 3R). Moreover, plasma HDL cholesterol concentration in mice depends on catechins' stereoisomerism and increases along with the increasing content of catechins belonging to the (-) form (2S, 3R).

Comparative matrix isolation infrared spectroscopy study of 1,3- and 1,4-diene monoterpenes ($\alpha$-Phellandrene and $\gamma$-Terpinene)

In the present work, $\gamma$-terpinene (a 1,4-diene derivative) and $\alpha$-phellandrene (1,3-diene derivative) were isolated in cryogenic argon matrices and their structures, vibrational spectra, and photochemistries were characterized with the aid of FTIR spectroscopy and quantum chemical calculations performed at the DFT/B3LYP/6-311++G(d,p) level of approximation. The molecules bear one conformationally relevant internal rotation axis, corresponding to the rotation of the isopropyl group. The calculations provide evidence of three minima on the potential energy surfaces of the studied molecules, where the isopropyl group assumes the trans, gauche+, and gauche conformations (T, G+, G- ). The signatures of all these conformers were identified in the experimental matrix infrared spectra, with the T forms dominating, in agreement with the theoretical predicted abundances in gas phase at room temperature. In situ UV ($\lambda$ > 200 nm) irradiation of matrix-isolated $\alpha$-phellandrene led to its isomerization into an open-ring species. The photoproduct was found to exhibit the ZE configuration of its backbone, which to be formed from the reactant molecule does not require extensive structural rearrangements of both the reagent and matrix. $\gamma$-Terpinene was photostable when subjected to irradiation under the same experimental conditions. In addition, the liquid compounds at room temperature were also investigated by FTIR-ATR and FT-Raman spectroscopies.
