Opis:: Highly immunogenic nucleotide fragments from 3 genes of Mycoplasma hyopneumoniae strain 232 were selected using information software technology. After repeating each fragment three times, a total of 9 nucleotide fragments were joined together to form a new nucleotide sequence called Mhp2321092bp. Mhp2321092bp was directly synthesized and cloned into a pET100 vector and expressed in Escherichia coli. After purification, the proteins were successfully validated by SDS-PAGE and Western blotting using mouse His-tag antibody and pig anti-Mhp serum. BALB/c mice were intraperitoneally injected with purified proteins in the high-dose (100 µg), medium-dose group (50 µg) and low-dose (10 µg) groups. Mice in each group were injected on day 1, day 8 and day 15 of feeding, respectively. Serum samples were collected from all mice on the day before immunization and on day 22 after immunization. The antibody level in the mouse serum was detected using western blotting using purified expressed proteins as antigens. IL-2, TNF-α and IFN-γ were simultaneously detected in the mouse serum by ELISA. The results showed that the 60 kDa protein was successfully expressed and reacted specifically with the specific serum Mhp His-Tag mouse monoclonal antibody and pig anti-Mhp serum. From day 0 to day 22 of immunization, IFN-γ increased from 269.52 to 467.74 pg/mL, IL-2 increased from 14.03 to 145.16 pg/mL, and TNF-α increased from 6.86 to 12.37 pg/mL. The IgG antibody in mice increased significantly from 0 day to day 22 after immunization. This study suggests that the expressed recombinant protein may serve as one of the novel vaccine candidates for Mhp.

Opis:: Tunnel thrusters during overhaul pass through several quality tests. After disassembly any clearance, shrink or subassembly are noticed. Measurements taken at verification stage determined which parts should be replaced. Definitely, the most important of tunnel thruster operation is bearings condition. Despite many years of experience, it is hard to find micro damages in the bearings without special devices. After assembly of tunnel thruster, the final stage is spin test. The spin test rig is equipped with bearings condition monitoring system including vibration, temperature and pressure inside the tunnel thruster live monitoring. During 4 hours of spin test, it is possible to check vibrations on the thruster and if any damage occurs it can be easily found, on which part of bearing appeared. Spin test rig gives possibility to check misalignments, gear condition, temperatures during 4 hours of operation and check oil quality (oil particle content acc. to ISO 4407 and water content). Spin test guarantees proper operation of tunnel thrusters after overhaul. If any damage appears during the spin test it can be easily eliminated. Without the spin test after overhaul, damage will be found after assembly on the vessel – on the sea trials. When damage is found too late, there is an additional cost for company to dock the vessel again and disassembly tunnel thruster for inspection. From economical point of view, the spin test rig gives possibility to avoid extra costs. Every passed spin test gives certificate of proper vibration level and proper assembly of thruster’s elements.

Opis:: This article presents the research results, which set out to explain the mechanisms leading to the achievement of dominance by the Russian Federation in the international security environment. In the research process, the systemic approach was applied. Analysis and criticism of the literature, non-participatory observation and case study elements were used to solve the research problems. As a result, it was determined that the theoretical basis for the Russian Federation's achievement of international dominance is the concept of new generation war. The model of the strategic influence of the Russian Federation is directed at the shaping of the security environment and includes the synchronisation of kinetic and non-kinetic measures, indirect and direct effects, the blurring of the boundaries between war and peace and the application of pressure and aggression. The Russian Federation achieves strategic dominance through asymmetry, chaos, reflexive control, and strategic deterrence. Armed forces provide a key role in asserting dominance, focused on conducting offensive activities and inflicting losses with conventional and nuclear weapons.

Opis:: The role of “portraits of words” card index in the compiling of new generation dictionariesThe article presents the concept of “portraits of words” card index as a necessary tool of the analysis of language changes for the compiling of dictionaries of new generation. Such card index contains information not only about the use of words in the texts of various functional styles, time, subject and purpose, but also about their systemic characteristics: paradigmatic, syntagmatic and epidigmatic (derivational and associative). The information about a word gathered in the card index of this type serves as guidelines for its verification in the corpus as well as in the various text databases, including online resources. It allows to check the use of new words, their spelling variations, development of their semantics, the course of their competition with other nominations of the same realities, processes or attributes in the texts. A “portraits of words” card index, as juxtaposed with the corpus and texts, signals the need to supplement the corpus with certain types of texts, as well as the need to change the annotation of its texts. This enables the preparation of verified materials for the compiling of new generation dictionaries. Rola kartoteki „portretów słów” w tworzeniu słowników nowej generacjiW artykule przedstawiono koncepcję kartoteki „portretów słów” jako narzędzia analizy zmian językowych niezbędnego przy tworzeniu słowników nowego pokolenia. Kartoteka taka mieści w sobie informacje nie tylko o tym, jak słowo używane jest w tekstach o różnych stylach funkcjonalnych, z różnych okresów, o różnej tematyce i różnym przeznaczeniu, lecz także o jego charakterystykach systemowych: paradygmatycznych, syntagmatycznych i epidygmatycznych (słowotwórczych i asocjacyjnych). Informacje na temat słowa zgromadzone w takiej kartotece będą służyć jako swego rodzaju punkty orientacyjne przy weryfikacji występowania danego wyrazu w korpusie i bazach tekstów, zwłaszcza w internecie. Informacje te dają możliwość przeprowadzania porównań z tekstami pod kątem występowania nowych słów, wariantów ich pisowni, rozwoju ich semantyki, przebiegu ich konkurencji z innymi sposobami opisu tych samych realiów, procesów czy cech. Kartoteka tego rodzaju, w zestawieniu z korpusem i tekstami, sygnalizuje konieczność uzupełnienia korpusu o pewne rodzaje tekstów, a także potrzebę zmiany oznaczeń umieszczonych w korpusie tekstów – a tym samym pozwala na przygotowanie zweryfikowanego materiału wyjściowego do tworzenia słowników nowego pokolenia.

Opis:: The actions that can mitigate the adverse effects of various energy crises are continuous improvements in the efficiency of power generation plants. This can be achieved by carrying out the processes at an always higher temperature and pressure, and in a more aggressive environment than the parameters used so far. Such requirements cannot be satisfied any longer by the iron-based alloys, even the best ones. Therefore, multi-component alloys based on nickel and cobalt start to be the materials of choice. On the other hand, new materials of higher performance life are searched for all the time. Jointly with its American partners, the Foundry Research Institute in Krakow carries out the research works which, among others, aim at the conversion of both material and techno-logy from structures forged and welded in nickel superalloys to cast elements for operation under the most demanding conditions of the power plants of a new generation. A part of the research is done under an “A-USC - NICKEL” American programme, to participation in which the Foundry Research Institute has been invited. Within this programme, the initial studies have already been carried out to master the technique of melting and casting the Inconel 740 alloy and preliminary material testing has been performed on the ready castings. It has been stated, among others, that with the temperature increase, particularly above 700°C, this alloy when used as a cast material is characterised by a definitely less drastic decrease of tensile strength than the same alloy subject to plastic forming.
Elementem mogącym łagodzić skutki różnych kryzysów energetycznych jest ciągłe zwiększanie sprawności urządzeń energetycznych. Można to uzyskać prowadząc procesy technologiczne w coraz wyższej temperaturze, ciśnieniu i agresywniejszym środowisku niż stosowane dotychczas. Takim wymogom nie mogą sprostać już stopy na bazie żelaza, nawet najlepsze z nich pod tym względem. Sięga się tu po wieloskładnikowe stopy na bazie niklu i kobaltu. Powstają wciąż nowe tworzywa o coraz większej trwałości eksploatacyjnej. Instytut Odlewnictwa w Krakowie wspólnie z partnerami amerykańskimi prowadzi prace badawcze, których celem jest między innymi dokonanie konwersji materiałowej i technologicznej konstrukcji kuto-spawanych wykonywanych z nadstopów niklu, na elementy odlewane z przeznaczeniem do pracy w najtrudniejszych warunkach eksploatacyjnych urządzeń energetycznych nowej generacji. Jest ona realizowana między innymi w ramach amerykańskiego programu „A-USC – NICKEL”, do prac, w którym został zaproszony Instytut Odlewnictwa. W ramach tych prac wykonano już wstępne badania polegające na opanowaniu techniki topienia i odlewania stopu Inconel 740 oraz przeprowadzono wstępne badania materiałowe wykonanych odlewów. Stwierdzono między innymi, że ze wzrostem temperatury, zwłaszcza powyżej 700°C, stop ten, jako stop odlewniczy wykazuje zdecydowanie mniejszy spadek wytrzymałości na rozciąganie niż analogiczny stop przerabiany plastycznie.

Opis:: The extensive development of social entrepreneurship transnationally is being increasingly shaped by youth entrepreneurs from the so-called millennial generation. The newest reports unveil that social entrepreneurship has become a youthful movement engaged in developing social initiatives worldwide. The article is an exploratory and pilot research with an objective to verify the profile of the new generation of social entrepreneurs. The author applies qualitative, narrative and cross case study analysis of the young social entrepreneurs (under the age 30) giving insights into 71 social ventures from 14 European countries and 5 overseas. The study focuses on e.g. innovation types, fields of operation, capacity of social impact, market orientation and revenue capacity models, founders’ biographical traits and their opportunity recognition patterns. The findings discover a few leading features within the members of the new generation of social entrepreneurs such as, inter alia, increasing market orientation, high utilization of technological solutions (especially in the area of social exclusion) or approximate level of pull and push factors that engage young entrepreneurs to create social enterprises.
Intensywny rozwój ekonomii społecznej jest coraz częściej kształtowany przez tzw. pokolenie millenialsów. Najnowsze raporty dotyczące przedsiębiorczości społecznej zauważają wzrost zaangażowania młodych przedsiębiorców w rozwój inicjatyw społecznych w skali globalnej. Artykuł podejmuje próbę weryfikacji kluczowych cech omawianego zjawiska oraz utworzenia profilu młodego przedsiębiorcy społecznego. W tym celu autorka analizuje studium przypadku wybranych przedsiębiorstw społecznych założonych przez młodych przedsiębiorców poniżej 30. roku życia. Próba obejmuje 71 podmiotów społecznych działających w Europe i na świecie. Badanie jakościowe dokonuje charakterystyki młodych przedsiębiorców społecznych według poszczególnych kategorii, m.in.: głównych typów innowacji społecznych, rodzaju wpływu społecznego i jego skali (globalna lub lokalna), orientacji rynkowej oraz specyficznych aspektów biograficznych i motywów działania w obszarze ekonomii społecznej. Badanie wykryło m.in. wzrost orientacji rynkowej, liczne technologiczne rozwiązania głównie w obszarze wykluczenia społecznego oraz zbliżone wartości czynników zachęcających i zmuszających (tzw. ‘pull/push factors’) mających wpływ na tworzenie się podmiotów ekonomii społecznej wśród młodych przedsiębiorców w wymiarze międzynarodowym.

Celem zawartych w pracy rozważań dokonanych na bazie analizy źródeł oraz przeprowadzonego badania własnego jest scharakteryzowanie owej kultury oraz zweryfikowanie jej odmienności względem innych festiwali, a także określenie jej związków z charakterem festiwalu, określanego mianem „festiwalu nowej generacji”. W obliczu głębszych zmian dotyczących wielu różnych dziedzin życia dochodzi do transformacji charakterystyki części współczesnych festiwali. W opinii autorki festiwale te, które zgodnie ze słowami organizatorów Transatlantyk Festival określić można mianem „festiwali nowej generacji”, różnią się od innych festiwali – „festiwali starej generacji” – kulturą organizacyjną odpowiedzialnych za ich organizację podmiotów. Przeprowadzone badanie zdaje się potwierdzać ową hipotezę, zwracając uwagę na bardziej elastyczny oraz otwarty na zmiany i większą różnorodność charakter kultury organizacyjnej festiwali nowej generacji związany ponadto z bardziej zespołowym charakterem pracy wewnątrz Fundacji „Transatlantyk Festival”.
The focus of this master's thesis is set upon film festival called Transatlantyk Festival and the organizational culture of its' organizational unit – The 'Transatlantyk Festival' Fundation. The aim of the author's deliberations based upon existing content analysis and research conducted within this thesis is to characterize the organizational culture of The 'Transatlantyk Festival' Fundation, verify whether or not it is different from other festivals, and define its relation towards festival's character, which is described as 'a new generation festival'. In a situation of deep changes that affect many different parts of life transformation of characteristics of some modern festivals occurs. In the author's opinion these festivals, which can be named 'new generation festivals' in accordance with the words of Transatlantyk Festival's organizators, are different from other festivals – 'old generation festivals' – in terms of organizational culture of their organizational units. Research conducted within this thesis seems to confirm this hypothesis by noting more elastic and open nature towards changes and bigger diversity of new generation festivals' organizational culture, which is also related to more collaborative way of working within The 'Transatlantyk Festival' Fundation.
