    Opis:
    The association of phytoplasma was investigated in sand olive [Dodonaea viscosa ssp. Angustifolia (L. f.) J.G. West], cowpea [Vigna unguiclata (L.)] Wap and alfalfa (Medicago sativa L.) plants exhibiting witches broom, fasciation and little leaf symptoms, respectively. Sequence analysis of ~1.7 kb DNA fragments amplified by P1/P7 primer set confirmed the association of ‘Candidatus Phytoplasma aurantifolia’ within symptomatic alfalfa, while ‘Ca. Phytoplasma cynodontis’ was associated within cowpea and sand olive.

    Opis:
    Roots of Gladiolus x hybridus Van Houtte plants infected with aster yellows phytoplasma were examined. The infected plants had a reduced root system in comparison to control plants. Their roots were thinner and the stele organisation was changed. Phytoplasmas were present in sieve tubes, companion cells and phloem parenchyma cells of the infected plant roots. Free calcium ions were localized in the cells of infected plants. Cells of the stele of infected roots, especially these infected with phytoplasmas, showed an increase of calcium antimonite deposits in theirs protoplasts. Also the number of calcium antimonite deposits increased in sieve tubes of infected roots. The deposits were present on plasma membrane, around the sieve tube plate and also in the lumen of the sieve tube. The increase of free calcium ions in sieve tubes did not cause the occlusion of sieve tube pores. Companion cells and some parenchyma cells with phytoplasmas did not react to phytoplasma infection with an increase of Ca2+ ions in protoplast. The parenchyma cells showing signs of degeneration reacted with high increase of calcium ions. The Ca2+ ions were present mainly in cytoplasm of infected parenchyma cells. There were calcium antimonite deposits in infected plant roots xylem elements and in intracellular spaces of cortex parenchyma. Such deposits were not present in control plants.

    In 2017 growing season numerous examinations of Cosmos bipinnatus in Hormozgan province, Iran revealed the disease symptoms similar to those associated with phytoplasmas. Phytoplasmas were detected from all symptomatic plants by the specific polymerase chain reaction (PCR) utilizing phytoplasma universal primer pairs. Amplification, sequencing and blast analysis of 16S rDNA fragment (ca. 1.2 kb) demonstrated that C. bipinnatus plants were infected by a phytoplasma belonging to the 16SrII group. This is the first report of association of a ‘Candidatus Phytoplasma aurantifolia’-related strain with C. bipinnatus phyllody in Iran.

    During 2015, samples from 22 apple trees showing proliferation symptoms were collected in southwest Bulgaria and Central and South Poland and tested for phytoplasma presence. ‘Candidatus Phytoplasma mali’ was identified in 18 samples based on results of restriction fragment length polymorphism (RFLP) analysis of the 16S rRNA gene amplified in nested PCR using primer pair P1/P7 followed by R16F2n/R16R2 and F1/B6 primer pairs. The nitroreductase and rhodonase-like genes and ribosomal protein genes rpl22 and rps3 were then analyzed using PCR-RFLP technique to study the genetic variability of the phytoplasma strains. Two restriction profiles, P-I or P-II, were obtained from fragments of 16S rDNA plus 16S-23S spacer region digested with HpaII enzyme. Restriction fragment length polymorphism analysis of nitroreductase and rhodonase-like genes using digestion with HincII endonuclease revealed that all ‘Ca. P. mali’ strains belonged to the subtype AP-15. Analysis of rpl22 and rps3 ribosomal protein genes digested with AluI enzyme resulted in classification of detected phytoplasma strains to rpX-A subgroup.

    W latach 1998-2005 prowadzono obserwacje i badania nad występowaniem fitoplazm w drzewach i krzewach ozdobnych w Polsce. Przedmiotem badań były krzewy z rodzaju Clematis sp., Magnolia spp. i Rosa sp. oraz drzewa z rodzaju Acer sp., Carpinus sp., Fagus sp., Fraxinus sp. i Quercus spp. wykazujące objawy chorobowe sugerujące porażenie przez fitoplazmy. Objawy te obejmowały zahamowanie wzrostu, zmiany w zabarwieniu a nawet nekrozę i deformację liści, proliferację i zamieranie pędów, czasem deformację i zielenienie lub brak kwiatów oraz zamieranie roślin. Do wykrywania i identyfikacji fitoplazm stosowano test biologiczny oparty na reakcji rośliny testowej Catharanthus roseus, zakażanej techniką szczepienia, oraz analizę PCR-RFLP. Stosując technikę szczepienia, fitoplazmy przeniesiono z chorych róży, magnolii i klonu na siewki C. roseus. Przy pomocy techniki łańcuchowej reakcji polimerazy (PCR) ze starterami amplifikującymi fragment 16Sr DNA fitoplazm, obecność fitoplazmatycznego DNA wykryto w roślinach Rosa spp., Magnolia spp., Clematis sp., Fraxinus excelsior, Carpinus sp. i Acer negundo oraz w eksperymentalnie zakażonych siewkach C. roseus. Analiza RFLP produktu amplifikacji z użyciem enzymów restrykcyjnych AluI, HhaII MseI i RsaI wykazała, że rośliny grabu, jesionu, klonu, magnolii, powojnika i róży były naturalnie porażone przez fitoplazmę z grupy żółtaczki astra (16SrI-B) - 'Ca. Phytoplasma asteris’. W niektórych roślinach róży i magnolii stwierdzono także fitoplazmę z grupy proliferacji jabłoni (16SrX-A) - ‘Ca. Phytoplasma mali’. W badanych roślinach fitoplazmy występowały nierównomiernie i w stosunkowo niskiej koncentracji, przez co mogły być wykrywalne dopiero w drugiej rundzie PCR. W roślinach buka i dębu, mimo wyraźnych symptomów, nie wykryto obecności fitoplazmatycznego DNA.
    The presence of phytoplasmas in six plant species (Acer sp., Carpinus sp., Fagus sp, Fraxinus sp. i Quercus spp) with severe symptoms of leaf discoloration and malformation, leaf and shoot necrosis, abnormal production of secondary shoots and decline, as well as in periwinkle plants experimentally inoculated by grafting was demonstrated by PCR with primer pairs amplifying phytoplasma 16Sr DNA fragment. The RFLP analysis of the PCR product done with restriction enzymes AluI, HhaII MseI and RsaI showed that the tested rose, magnolia, clematis, ash-leaf maple, ash and hornbeam plants were naturally infected with aster yellows phytoplasma (16SrI-B), now reclassified as ‘Candidatus Phytoplasma asteris’. Apple proliferation phytoplasma (16SrX-A), now reclassified as 'Candidatus Phytoplasma mali’, was detected in diseased roses and magnolias. The phytoplasmas were stated using nested PCR in affected woody plants, indicating their law titre and uneven distribution. We failed to detect the phytoplasma presence in diseased Quercus and Fagus trees.

    In two of Iran's central provinces, several herbaceous plants showing phytoplasma disease symptoms were collected to detect 'Canididatus Phytoplasma asteris'-related phytoplasmas. Confirmation of an association of phytoplasmas with diseased plants was done using polymerase chain reaction (PCR) assays having the phytoplasma universal primer pairs P1/P7 followed by R16F2n/ R16R2 in nested PCR. Then, for detection of 'Ca. P. asteris', DNA samples were subjected to amplification of rp and tuf genes using specific primer pairs rp(I)F1A/rp(I)R1A and fTufAy/rTufAy, respectively. Restriction fragment length polymorphism or RFLP analyses of rp gene fragments using Tsp509I restriction enzyme as well as sequence analyses indicated that 'Ca. P. asteris'-related phytoplasmas associated with carrot, niger seed and scallion plants in these regions, belong to the rpI-L subgroup. This research is the first report of carrot, niger seed, and scallion infection with phytoplasmas belonging to the rpI-L subgroup.

    Recently, Russian olive trees showing witches’-broom and little leaf symptoms have been widely observed in northwestern and central Iran. Polymerase chain reaction (PCR) and nested PCR assays using phytoplasma universal primer pairs confirmed phytoplasma symptomatic infection of trees. Sequence analyses showed that ‘Candidatus Phytoplasma asteris’ was the causal agent of the disease in these regions. However, RFLP results using restriction enzymes HpaII, EcoRI, HinfI and AluI indicated that the collected isolates in these regions are genetically different. In addition, leafhopper Macropsis infuscata was recognized as a possible insect vector of the disease for the first time.

    The results of PCR examination indicated that during two years of tissue culture at standard conditions, on the medium with BAP 1 mg l-1 and continuous temperature of 20oC, phytoplasma could be detected in diseased plants of rose cv Sacha and Jazz. In the second year of micropropagation phytoplasma detection rate in tissues of infected roses increased and was relatively higher than in the first one. To test whether phytoplasmas are sensitive to temperature and light intensity, phytoplasma-affected micropropagated rose plants were grown on medium with BAP 1.0 or 0.5 mg l-1 and at the temperature of 4, 15, 20 or 25oC in darkness or in the light. PCR analysis indicated that phytoplasma detection was not effected by these conditions during 4 weeks of culturing. However, phytoplasma was not detectable in rose plants after 8 weeks culturing on the same medium without transplanting. Micropropagated rose shoots maintained on medium with Gentamycin or Baytril at the concentration of 25.0 or 50.0 mg l-1 had reduced growth and were chlorotic. However, no direct effect of applied antibiotics on phytoplasma detection was evidenced.

    In the spring of 2012, sophora (Sophora alopecuroides L.) plants showing symptoms of leaf yellowing, little leaves and stunting were observed in Firooz-kuh (Tehran province), Sari (Mazandaran province) and Urmia (West Azerbaijan province) in Iran. Symptomatic plants from the three locations were subjected to nested polymerase chain reaction (PCR) to amplify 16SrRNA using primer pair P1/P7 followed by primer pair R16F2n/R16R2. Th e amplicons were purifi ed, sequenced and the nucleotide sequences were analyzed by virtual restriction fragment length polymorphism (RFLP). Th e phytoplasmas associated with the yellows disease were identifi ed as members of the 16SrIX group (Candidatus Phytoplasma phoenicium) and the 16SrXII group (Candidatus Phytoplasma solani). Th e two phytoplasmas were placed in 16SrIX-C and 16SrXII-A subgroups, respectively, in constructed phylogenetic trees. Th is is the fi rst report on sophora yellows associated with Candidatus Phytoplasma phoenicium.
