Opis:
Rebaudioside A (RebA) is a steviol glycoside used for production of sweeteners. It was shown that the glycosides affect memory and learning processes. The aim of the study was to investigate neurons immunoreactive for acetylcholinesterase (AChE) and M1 muscarinic receptors (mAChRs-M1) of the hippocampal CA1 and CA3 fields and striatal caudateputamen (CP) and globus pallidus (GP) in rats receiving RebA. RebA was administrated to adult rats for 45 days in dilutions of 1 mg and 2 mg RebA/ml water. Indirect peroxidase-antiperoxidase immunohistochemical reaction was conducted on frontal sections containing the hippocampus and striatum with use of antibodies against AChE and mAChRs-M1. Immunoreactive for the studied proteins neurons were morphologically and morphometrically assessed in hippocampal CA1 and CA3 fields and in CP and GP. Microscopic observations did not reveal significant changes in morphology of immunoreactive neurons, which suggests no neurotoxic effect of the studied glycoside on these cells. Morphometric analyses revealed an increase in the density of AChE and mAChRs-M1 immunoreactive neurons. A decrease in reaction intensity of AChE-positive neurons was also demonstrated in the hippocampal CA1 field and in GP. In contrast, an increase in reaction intensity of mAChRs-M1-positive neurons was found in CA1, CA3 fields and in CP and GP. The results of our preliminary studies indicate that RebA administrated to rats has an impact on cholinergic neurons in the studied area. The results suggest a possible increase in the activity of the cholinergic system, responsible for memory and learning processes, after administration of RebA.

Stevia rebaudiana Bertoni is a desirable sugar substitute due to the presence of natural sweeteners (steviol glycosides). In 2011 steviol glycosides from this plant were accepted by the European Food Safety Authority (EFSA) as a harmless sweetener added to foods.The plant contains 9 different steviol derivatives commonly referred to as steviol glycosides. The largest share among them are: stevioside and rebaudioside A (the main components responsible for the sweet taste of the plant), while the others are rebaudioside B, C, D, E, F, dulcoside A and B and steviolbioside. The highest concentration of these substances is observed in the leaves. Stevia leaves may differ in the content of steviosides depending on the source. Therefore, the aim of the study was to compare the content of steviosides in stevia leaves (Stevia rebaudiana) from different countries and in products produced on the basis of this plant. On the basis of stevia leaf analysis, it was shown that the content of steviol glycosides in stevia leaves - rebaudioside A and stevioside, depends on the place and conditions in which it is grown. In addition, the amount of these substances in products based on this plant was analyzed. The study showed that they can be a substitute for dried leaves provided that the recommended daily intake is not exceeded.

Seeds of $\textit{Stevia rebaudiana}$ Bertoni as a source of plant growth-promoting endophytic bacteria with the potential to synthesize rebaudioside A

In this study, a new strain of Pantoea vagans, SRS89, was isolated from surface-sterilized stevia seeds. The isolate was evaluated using morphological, molecular, and biochemical methods. The bacterium was 1.5 μm long, yellowish in color, and classified as Gram-negative. Whole genome sequencing of our strain revealed the presence of a 4,610,019 bp chromosome, and genome annotation resulted in the detection of 4283 genes encoding 4204 putative coding sequences. Phylogenic analysis classified the genome of our strain close to the MP7 and LMG 24199 strains of P. vagans. Functional analysis showed that the highest number of genes within the analyzed bacterium genome were involved in transcription, amino acid transport and metabolism, and carbohydrate transport and metabolism. We also identified genes for enzymes involved in the biosynthesis of carotenoids and terpenoids. Furthermore, we showed the presence of growth regulators, with the highest amount noted for gibberellic acid A3, indole-3-acetic acid, and benzoic acid. However, the most promising property of this strain is its ability to synthesize rebaudioside A; the estimated amount quantified using reversed-phase (RP)-HPLC was 4.39 mg/g of the dry weight of the bacteria culture. The isolated endophytic bacterium may be an interesting new approach to the production of this valuable metabolite.

The separation and purification of rebaudioside A from Stevia rebaudiana crude extracts (Steviosides) by macroporous resin were optimized by Taguchi orthogonal array (OA) experimental design methodology. This approach was applied to evaluate the influence of five factors (adsorption temperature, desorption time, elution solution ratio, adsorption volume and type of resin) on the rebaudioside A yield. The percentage contribution of each factor was also determined. The results showed that elution solution ratio and adsorption volume made the greatest (59.6%) and the lowest (1.3%) contribution, respectively. The results showed that the Taguchi method is able to model the purification of rebaudioside A process well (R2 > 0.998) and can therefore be applied in future studies conducted in various fields. Adsorption temperature 35°C, desorption time 60min, elution solution ratio 3, adsorption volume 200ml and HPD-400 as resin were the best conditions determined by the Taguchi method.

The final results of this study was development of a new, simple, rapid, high throughput and cheap high performance thin layer chromatographic technique with densitometric detection for identification and quantification of two steviol glycosides. Analitycal procedure presented in this thesis has been successfully validated. The selectivity, precision and accuracy was confirmed. There has been established the range of the method, detection limit and limit of quantitation. A curvilinear relationship was found in this method.Development of a new mobile phase enable optimization of method conditions and to achieve a better value of retention factor Rf. The method was characterized by simple, one-step extraction process and detection in UV range. The assays has not been interfere with any other substances that occurred in analyzed samples. Direct analysis was performed without the need of derivatization.It was the first time identification and quantitative determination of stevioside and rebaudioside A in sweeteners. Researches revealed differences in purification, composition of excipients and the contents of the main active components: stevioside and rebaudioside A in 5 sweeteners. The amount of rebaudioside A was proved to be greater than stevioside. Various properties of tablets was also demonstrate based on different disintegration time.In conclusion, a new method was found to be reliable in this examination and could be used for the routine analysis of steviol glycoside. This is especially important becauce of the fact that Stevia rebaudiana (Bertoni) is in increasingly common use.
Ostatecznym wynikiem pracy było opracowanie nowej, prostej, szybkiej, wysokowydajnej i taniej metody HPTLC z wykorzystaniem detekcji densytometrycznej, do identyfikacji i ilościowego oznaczania dwóch glikozydów stewiolowych. Metoda została pomyślnie zwalidowana, potwierdzono jej selektywność, precyzję oraz dokładność. Wyznaczono zakres metody, granicę wykrywalności oraz oznaczalności. Stwierdzono krzywoliniowość wskazań w zakresie metody.Opracowanie nowej fazy rozwijającej pozwoliło na optymalizację warunków metody i osiągnięcie korzystniejszych wartości współczynnika opóźnienia Rf. Metoda odznacza się również prostym, jednoetapowym procesem ekstrakcji badanych związków oraz umożliwia detekcję w zakresie UV, dzięki czemu nie ma konieczności wybarwiania otrzymanych chromatogramów. Analiza została przeprowadzona w sposób bezpośredni. Nie stwierdzono występowania interferencji ze składnikami współobecnymi.W przebiegu analizy zidentyfikowano i ilościowo oznaczono stewiozyd i rebaudiozyd A w słodzikach, co stanowi pierwsze jak do tej pory badanie w tego rodzaju materiale. Badane słodziki różniły się stopniem oczyszczenia, składem substancji pomocniczych a co najważniejsze zawartością składników głównych, czyli stewiozydu i rebaudiozydu A. Ilość rebaudiozydu A okazała się być większa niż stewiozydu. Również przeprowadzone badanie czasu rozpadu tabletek poszczególnych słodzików, ukazujące różną szybkość rozpuszczania, pozwoliło na stwierdzenie ich odmiennych właściwości.Podsumowując, opracowana metoda okazała się być przydatna w przeprowadzonym oznaczeniu i może być wykorzystana w rutynowych analizach glikozydów stewiolowych. Jest to szczególnie ważne ze względu na fakt, iż Stevia rebaudiana (Bertoni) jest w coraz powszechniejszym użyciu.

Stevia rebaudiana Bertoni is famous for sweetness due to the content of steviol glycosides (SG). This study aimed to determine the effect of SG (stevioside, rebaudioside A) and steviol on adipogenesis and lipogenesis in the 3T3- L1 model. Test compounds were also analysed for their ability to affect glucose uptake in hypertrophied insulin- resistant 3T3-L1 adipocytes. The most active compound in modulating adipogenesis, lipogenesis and insulin resistance was steviol, SG metabolism product. Steviol (10 μM, 100 μM) significantly down-regulated the expression of adipogenic transcription factors (PPARγ, C/EBPα, SREBP1) and lipogenic genes (FAS, aP2, LPL), which caused decreased lipid accumulation and triglyceride content in adipocytes. Treatment of insulin-resistant adipocytes with steviol (1 μM) and stevioside (1 μM, 10 μM) increased GLUT-4 transcript level and improved glucose uptake. Steviol also lowered resistin gene expression, which may mitigate insulin resistance in hyper- trophied adipocytes. This study promotes SG as potential health-promoting ingredients in low-calorie functional foods.

Stevia rebaudiana Bertoni which has gained industrial and scientific interests is a suitable nutritional alternative to sucrose as a sweetener. Recently, there have been studies which show the extraction of this phytochemical substance from stevia leaves and purification methods by several alcohols and chromatographic methods. However, these methods are not cost-effective. Therefore, an attempt was made to extract and purify ST using inexpensive, scalable and simple techniques where different steps like extraction, electrocoagulation, ion exchange, activated charcoal, vacuum evaporation and butanol wash were used as purification steps. The present study established a new improved technology of extraction of ST from stevia leaves using water as a solvent followed by various purification steps. 496 mg of Stevioside extracted in the form of crystals was obtained from 100 g of leaves which is 10 times more than the reported yield of 54 mg from 100 g stevia leaves in literature. This methodology can be scaled up at the industry level for future large production to meet the huge demand for natural sweeteners.
