Opis:
The aims of our study were to assess the release of cytotoxic nucleoside analogs 5-fluorouracil and 2-chloro-2'-deoxyadenosine from different lactide-glycolide or lactide-caprolactone biodegradable copolymers and the effects of sterilization on this release. The polymers were sterilized either with ethylene oxide at 37°C, or with gamma radiation (15 kGy, 20 kGy, or 25 kGy). The kinetics of nucleoside release from the copolymers were measured over 50 days. Four copolymers exhibited relatively constant release of nucleosides in micromolar concentrations during the entire observation period. Sterilization with either ethylene oxide or gamma radiation only slightly influenced nucleoside release. Further development of these copolymers as an intracerebral nucleoside delivery system for local treatment of brain tumors is indicated.

Opis:
Preparation of scaffolds for tissue regeneration requires elucidation of chemical and physical processes induced by ionizing radiation during radiation sterilization of polymeric materials used for these purposes. Such treatment induces many unintended and desired reactions that lead either to degradation or cross-linking of macromolecules. It was found that in poly(ester urethane) the ester segments are responsible predominantly for radiation induced phenomena. Some radicals generated following irradiation were identified under cryogenic conditions. Upon exposure to sterilizing dose cross-linking was confirmed on the basis of rheological changes in melted polymers. The results obtained by EPR spectroscopy and DRS revealed a relatively high susceptibility of poly(ester urethanes) on oxidation.

Opis:
The influence of ionising radiation on physicochemical properties of betamipron in solid state was studied. No changes for betamipron irradiated with a dose of 25 kGy, required to attain sterility, was observed by spectroscopic and chromatographic methods. Solid betamipron has proven to be very stable on irradiation, and irradiation has been found to be a suitable method for its sterilization.

The popular pharmaceutical base used in pharmacy – vaselinum flavum – was studied by an X-band (9.3 GHz) EPR spectrometer in the range of microwave power of 2.2–70 mW. The samples were sterilized in hot air oven at temperatures: 160°C (120 min), 170°C (60 min), and 180°C (30 min). The aim of this work was to determine properties and free radical concentrations in vaselinum flavum thermally sterilized at different conditions. The changes in free radical system in vaselinum flavum during storage were analyzed. Free radicals were found in all the heated samples. The lowest free radical concentration was obtained for vaselinum flavum heated at 180°C for 30 min; so these parameters are proposed for the thermal sterilization of this pharmaceutical base. Interactions with oxygen decreased free radical concentration in vaselinum flavum during storage. Strong quenching of free radicals in vaselinum flavum was observed after 2 days for the samples sterilized at temperatures 160 and 180°C. Such an effect for vaselinum flavum heated at temperature 170°C was observed later, 13 days after sterilization. Fast spin-lattice relaxation processes exist in thermally sterilized vaselinum flavum. The EPR lines of heated vaselinum flavum were homogeneously broadened. EPR spectroscopy and its use for examining the thermal sterilization process in pharmacy was confirmed.

The influence of ionising radiation on physicochemical properties of doripenem in solid state was studied. No changes for doripenem irradiated with a dose of 25 kGy, required to attain sterility, was observed by FT-IR and chromatographic methods, although the presence of free radicals (7.7*1017 radicals/g) was detected by EPR. The antibacterial activity of doripenem irradiated with a dose of 25 kGy was unchanged. Solid doripenem has proven to be very stable on irradiation, and irradiation has been found to be a suitable method for its sterilization.
