Opis:
Nowadays, Hepatitis B X interacting protein (HBXIP) is an object of scientists’ interest worldwide. It is a protein with significant involvement in the development of malignant tumors like breast or ovarian cancer. One of the most important functions of HBXIP is the regulation of cell proliferation, which is related to the progression of a cell cycle. Many studies provide the growing number of evidence that HBXIP plays various important roles, including the regulation of a cell cycle through complexes with survivin, belonging to the inhibitors of apoptosis and interactions with transcriptional factors like STAT4, SP1, TFIID or E2F1. It also has the influence on the promotion of tumor angiogenesis thanks to the association with VEGF and FGF8. Another important role of HBXIP is a reprogramming of glucose metabolism to conditions favorable to growing cancerous cells due to regulating the activation of SCO2 and PDHA1. Furthermore, it impacts on the complement-dependent cytotoxicity, also, HBXIP affects on lipid metabolism through disturbing of metabolic pathways of FAS. According to recent studies, HBXIP can be used as a prognostic biomarker in many tumors, including cervical cancer, ovarian cancer, and esophageal squamous cell carcinoma thanks to the high expression of this protein noted exclusively in these tumor tissues. What is even more interesting, it significantly correlates with clinical attributes like metastasis to lymph nodes or grading and in some cases can potentially be used as the indicator of prognosis of treatment effectiveness. The paper is review through main functions of HBXIP and its possible applications.

Multivariate statistical analysis, through their ability to extract hidden relationship between various traits, has a wide application in breeding programs. Having physiological concept on the multivariate analysis, factor analysis was used to extract differential relationships between different components involving in assimilate partitioning in sesame under regular irrigation regime and limited irrigation. The analysis revealed that under regular irrigation regime, the stored and/or currently produced assimilates are allocated to the filling seeds. However, incidence of water shortage in the beginning of flowering time make shifts in assimilate partitioning from formation of new seeds or capsules to the not-matured pre-formed seeds, which results in seeds with more nutrient storage. This indicates the requirement for change in breeding strategies under sub-optimal condition. The possible common language between factor concept in multivariate analysis, QTLs in genetics, and transcription factors in molecular biology is indicated.

Drought conditions may have direct or indirect effects on plant physiology, biochemistry, and molecular characteristics. The purpose of this study was to investigate the effects of drought stress on the physiological, biochemical, and molecular responses of three different watermelon cultivars with varying levels of drought tolerance (24: drought resistant, CS: moderately tolerant, and 98: drought sensitive). The cultivars exhibited different responses to cope with water stress according to their tolerance level. Drought induced significant reductions in chlorophyll a, total chlorophyll and carotenoid content and glutation reductase and ascorbate peroxidase activity in the sensitive cultivar unlike in the moderately tolerant and drought resistant cultivars. Additionally, the expression levels of NAC1, NAC2, ORE1, WRKY24, SAG12, SAG13, KCS2, CER1, DREB2A, LTP3, SWEET15, and PYL9 genes were measured using qRT-PCR. The expression ratios of the genes significantly varied depending on the gene location and on the tolerance of the cultivars. Results showed that the physiology and biochemical and molecular pathways of tolerant cultivars change to adapt to drought conditions. Therefore, the drought-resistant cultivar copes with drought stress by increasing proline content and antioxidant enzyme activities, as well as by increasing the expression of specific genes.

Opis:
Peganum harmala is a valuable wild plant that grows and survives under adverse conditions and produces pharmaceutical alkaloid metabolites. Using different assemblers to develop a transcriptome improves the quality of assembled transcriptome. In this study, a concrete and accurate method for detecting stress-responsive transcripts by comparing stress-related gene ontology (GO) terms and public domains was designed. An integrated transcriptome for P. harmala including 42 656 coding sequences was created by merging de novo assembled transcriptomes. Around 35 000 transcripts were annotated with more than 90% resemblance to three closely related species of Citrus, which confirmed the robustness of the assembled transcriptome; 4853 stress responsive transcripts were identified. CYP82 involved in alkaloid biosynthesis showed a higher number of transcripts in P. harmala than in other plants, indicating its diverse alkaloid biosynthesis attributes. Transcription factors (TFs) and regulatory elements with 3887 transcripts comprised 9% of the transcriptome. Among the TFs of the integrated transcriptome, cystein2/histidine2 (C2H2) and WD40 repeat families were the most abundant. The Kyoto Encyclopedia of Genes and Genomes (KEGG) MAPK (mitogen-activated protein kinase) signaling map and the plant hormone signal transduction map showed the highest assigned genes to these pathways, suggesting their potential stress resistance. The P. harmala whole-transcriptome survey provides important resources and paves the way for functional and comparative genomic studies on this plant to discover stress-tolerance-related markers and response mechanisms in stress physiology, phytochemistry, ecology, biodiversity, and evolution. P. harmala can be a potential model for studying adverse environmental cues and metabolite biosynthesis and a major source for the production of various alkaloids.

Opis:
MADS-box genes encode transcription factors in all eukaryotic organisms thus far studied. Plant MADS-box proteins contain a DNA-binding (M), an intervening (I), a Keratin-like (K) and a C-terminal C-domain, thus plant MADS-box proteins are of the MIKC type. In higher plants most of the well-characterized genes are involved in floral development. They control the transition from vegetative to generative growth and determine inflorescence meristem identity. They specify floral organ identity as outlined in the ABC model of floral development. Moreover, in Antirrhinum majus the MADS-box gene products DEF/GLO and PLE control cell proliferation in the developing flower bud. In this species the DEF/GLO and the SQUA proteins form a ternary complex which determines the overall "Bauplan" of the flower. Phylogenetic reconstructions of MADS-box sequences obtained from ferns, gymnosperms and higher eudicots reveal that, although ferns possess already MIKC type genes, these are not orthologous to the well characterized MADS-box genes from gymnosperms or angiosperms. Putative orthologs of floral homeotic B- and C-function genes have been identified in different gymnosperms suggesting that these genes evolved some 300-400 million years ago. Both gymnosperms and angiosperms also contain a hitherto unknown sister clade of the B-genes, which we termed Bsister. A novel hypothesis will be described suggesting that B and Bsister might be involved in sex determination of male and female reproductive organs, respectively.

Introduction and aim. Paired box 8 (PAX-8) is a specific transcription factor known as a protein product gene that plays an essential role in organogenesis and oncogenesis. The aim of this paper was to discuss structure and function of PAX-8. The aim of this study is to determine the utility of PAX-8 in cytology effusions with metastatic tumor. Material and methods. This article is a review done in regards to discuss the role knowledge on PAX-8 especially in oncogenesis and organogenesis. Analysis of the literature. Current information about PAX-8 is presented. Conclusion. The PAX family of genes plays an important role in the formation of tissues and organs during embryonic development and in maintaining the proper functioning of certain cells after birth.
