Opis:
A trehalose synthase gene from Deinococcus radiodurans (DSMZ 20539) containing 1659 bp reading frame encoding 552 amino acids was amplified using PCR. The gene was finally ligated into pET30Ek/LIC vector and expressed after isopropyl β-d-thiogalactopyranoside induction in Escherichia coli (DE3) Rosetta pLysS. The recombinant trehalose synthase (DraTreS) containing a His6-tag at the C-terminus was purified by metal affinity chromatography and characterized. The expressed enzyme is a homodimer with molecular mass of 126.9 kDa and exhibits the highest activity of 11.35 U/mg at pH 7.6 and at 30°C. DraTreS activity was almost unchanged after 2 h preincubation at 45°C and pH 7.6, and retained about 56% of maximal value after 8 h incubation at 50°C. The DraTreS was strongly inhibited by Cu2+, Hg2+, Zn2+, Al3+ and 10 mM Tris. The Km value of maltose conversion was 290.7 mM.

Opis:
Two recombinant trehalose synthases from Deinococcus geothermalis (DSMZ 11300) were compared. A significant influence of the artificial polyhistidine tag was observed in protein constitution. The recombinant trehalose synthase from D. geothermalis with His6-tag has a higher Km value of 254 mM, in comparison with the wild-type trehalose synthase (Km 170 mM), and displayed a lower activity of maltose conversion when compared to the wild type. Moreover, differences in properties like temperature, pH, thermal- and pH-stability were observed. Presence of the histidine tag caused a decrease of thermal resistance in case of trehalose synthase with His6-tag. These data confirmed a suggestion that the introduction of the histidine domain produces in some seldom cases undesirable changes in the protein.

α,α-Trehaloza jest naturalnie występującym disacharydem, zbudowanym z dwóch cząsteczek glukozy połączonych wiązaniem α,α- 1,1’-O-glikozydowym. Znana jest głównie ze zdolności do stabilizacji białek w warunkach stresu środowiskowego oraz hamowania ich agregacji. Po opracowaniu wielkotonażowej, biotechnologicznej metody produkcji, trehaloza znalazła liczne zastosowania w różnych gałęziach przemysłu, głównie w przemyśle spożywczym, kosmetycznym, farmaceutycznym i medycynie. Dowiedziono, że właściwości bioprotekcyjne trehalozy zostają zintensyfikowane w przypadku jej glikopolimerów.
α,α -Trehalose is naturally occurring disaccharide composed of two glucose molecules linked by α,α -1,1’-O-glycosydic bond. It is primarily known from its ability to protect proteins during environmental stresses and inhibition of their aggregation. The development of largescale bioprocess has allowed to use trehalose in various applications, mainly in the food, cosmetic and pharmaceutical industry as well as in medicine. It was reported that bioprotective properties are amplified when trehalose is incorporated in polymer chain as a pendant group.

Opis:
The dairy propionibacteria, which are traditionally used for the production of Swiss cheeses, are able to synthesize valuable biomolecules, e.g. B group vitamins, propionic acid, and trehalose with unique chemical and physical properties. Both, dairy propionibacteria cells and trehalose, have found many applications as attractive and effective components in food, beauty and health care products. This study confirmed the ability of several strains from the Propionibacterium genus to create trehalose from glycerol. The research aimed to investigate the effect of crude and pure glycerol on biomass production and on trehalose accumulation by Propionibacterium freudenreichii ssp. shermanii 1. The results indicated that the capacity for trehalose accumulation by Propionibacterium spp. was strain dependent. Propionibacterium freudenreichii ssp. shermanii 1 was able to grow on crude glycerol. For both, pure and crude glycerol, the highest amount of dry biomass leveled off at about 4 g/L. While the use of crude glycerol had no effect on the final concentration of biomass, it reduced the accumulation of trehalose in the cells. An increase in the concentration of carbon source (2-8%) resulted in more than a 5-fold rise in trehalose production. The highest trehalose concentration of 195.04 mg/L was obtained with cultures of the said strain supplemented to 8% with pure glycerol.

Celem badań było określenie wpływu zróżnicowanego dodatku dwucu-kru trehalozy do ciasta pszennego na cechy sensoryczne i fizykochemiczne bułek kajzerek. Dodatek trehalozy nie spowodował istotnego pogorszenia cech sensorycznych i fizykochemicznych produktu. Bułki kajzerki z dodatkiem trehalozy w ilości 0,5 i 1% w stosunku do mąki cechowały się dobrą jakością.
The aim of experiments was to investigate an influence of the additi of disaccharide trehalose to wheat dough on sensory and physicocl mical properties of Kaiser rolls. It was stated that the addition oft halose did not worsen sensory and phvsicochemical properties of t product. Kaiser rolls with the addition of 0.5 and 1% trehalose in p portion to flour were characterized by good quality.

Honey bees are resilient to the long-term presence of alcohol in their diet

Previous studies on various organisms have suggested that low doses of ethanol can have stimulatory effects, while higher doses may lead to toxicity, a response known as hormesis. Low ethanol concentrations occur naturally in the environment, particularly in fermenting fruits and flower nectar, where pollinators such as honey bees may encounter it. This study aimed to investigate the potential hormetic effects of low-level ethanol consumption on honey bees. Bees were divided into three groups: one provided with only sucrose solution, one both with sucrose and 0.5% ethanol in sucrose, and one with only 1% ethanol in sucrose. The bees were exposed to these diets for 14 days, and their performance was assessed through survivorship, flight endurance, body mass, lipid content, and trehalose and ethanol levels in the haemolymph. The results showed no significant differences in most parameters between the groups. However, bees constantly exposed to 1% ethanol had slightly higher trehalose levels compared to the control group, suggesting a possible adaptive response to ethanol exposure. Ethanol levels in the haemolymph differed significantly between groups, with bees exposed to ethanol showing detectable levels in their system. While no clear hormetic effects were observed in terms of improved performance, the elevated trehalose levels in bees constantly exposed to 1% ethanol may indicate adaptations protecting bees from ethanol-induced damage. The study provides insights into how honey bees tolerate low-level ethanol exposure and highlights the need for further research on the ecological implications of ethanol consumption in pollinators.

Recently, trehalose becomes more and more popular compound. Increasing interest in this disaccharide results from the role as it starts to play in water economy of plants. In conditions of water shortage mentioned organic substance prevents their dehydration. Therefore, application of such interesting additive can contribute to the preparation of materials that can be applied in such areas as agriculture or cultivation of plants. It is also worth mentioning that properties of trehalose make this substance interesting from the medical point of view. Presented additive can have an impact on maintaining organs for transplantation in good condition. In presented research series of polymer superabsorbents modified with described disaccharide have been prepared. Obtained by means of photopolymerization materials differed in an amount of the additive. In the further step, physicochemical properties of superabsorbents have been determined. Following studies have been carried out: swelling studies and incubation studies in simulated body fluids aimed at determination of behavior of prepared materials in solutions similar to those one occurring in human body. Additionally, chemical structure of the hydrogels have been defined using spectroscopic technique. Based on the research it can be concluded that proposed materials are characterized by low sorption capacity and did not have significant impact on simulated body fluids. What is more, prepared hydrogels did not degrade significantly in the mentioned fluids - such conclusion is possible due to the fact that spectra of the tested materials before and after incubation studies obtained using FT-IR spectroscopy differed slightly from each other. Presented superabsorbents seem to be interesting materials that can be subjected to more advanced studies in view of their future medical application.

The aim of the research was to check if trehalose and trehalose 6-phosphate affect velocity and amplitude of accumulation and avoidance responses of chloroplasts induced by blue light. Exogenous treatment with both substances does not show any influence on photometric parameters of the responses in Arabidopsis thaliana mesophyll cells. Additionally, experiments on inhibitory effect of 3% glucose have been repeated, in order to check whether type of soil and light conditions during plant growth influence that effect.
Przeprowadzono pilotowe doświadczenia mające na celu sprawdzenie czy trehaloza i trehalozo 6-fosforan mają wpływ na szybkość i amplitudę reakcji akumulacji oraz ucieczki w przemieszczeniach chloroplastów wywołanych światłem niebieskim. Wykazano, że obie substancje podane egzogennie nie wpływają na wyznaczone fotometrycznie parametry reakcji w mezofilu liści Arabidopsis thaliana. Dodatkowo powtórzono doświadczenia nad hamującym działaniem 3% glukozy na ruchy chloroplastów, celem sprawdzenia czy rodzaj podłoża i warunki świetlne podczas wzrostu roślin wpływają na ten efekt.
