Opis:: New psychoactive substances (NPS) sometimes referred to as “legal highs” or “designer drugs” are psychotropic materials that mimic the effects of other banned illicit drugs but are not always controlled under drug legislation. NPS are typically marketed as “not for human consumption” and are instead labelled and sold as research chemicals in Poland to avoid legislative controls. One of the main NPS classes are hallucinogens. The present paper aims at providing an overview of two groups of psychedelic drugs – lysergamides and tryptamines, and determining their legislation status in Poland. Psychodysleptics have always been of interest to people who want to experiment with their senses and boundaries. They may show beneficial effects which, from a medical point of view indicate the possibility of using them in therapies. NPS that have been described in this paper: 1-Propionyl-d-lysergic acid diethylamide (1P-LSD), 6-Allyl-6-nor-lysergic acid diethylamide (AL-LAD), 6-Ethyl-6-nor-lysergic acid diethylamide (ETH-LAD), 4-hydroxy-N-methyl-N-ethyltryptamine (4-HO-MET), 5-methoxy-N,N-diallyltryptamine (5-MeO-DALT).

Tytuł:: Developing of a fast and simple method of qualitative analysis for new psychoactive substances
Opracowanie szybkiej metody identyfikacji substancji zawartych w próbkach zabezpieczonych z rynku narkotykowego, tzw. dopalaczach i preparatach farmaceutycznych

Opis:: Niniejsza praca dotyczy opracowania szybkiej i prostej metody identyfikacji nowych substancji psychoaktywnych z wykorzystaniem techniki LC-MS/MS przy użyciu chromatografu cieczowego SCIEX Exion LC z autosamplerem próbek ciekłych sprzężonego ze spektrometrem mas z potrójnym analizatorem kwadrupolowym SCIEX Triple Quad 3500.W części teoretycznej w pierwszej kolejności opisano nowe substancje psychoaktywne, ich klasyfikację, regulacje prawne, podano również statystyki europejskie i polskie oraz bardziej szczegółowo opisano grupy tryptamin, syntetycznych katynonów i kannabinoidów, które badano w niniejszej pracy. Następnie skupiono się na metodach stosowanych do rozdziału oraz analizy jakościowej związków, czyli chromatografii cieczowej, spektrometrii mas i połączeniu tych obu technik, czyli LC-MS/MS. Na końcu opisano przykłady zastosowań tej metody w naukach sądowych, w szczególności do wykrywania NPS.W części doświadczalnej najpierw opisano cel badań, wykorzystywaną aparaturę, użyte odczynniki i sposób przygotowywania roztworów do badań. Następnie przedstawiono proces optymalizacji spektrometru mas, podczas którego udało się zoptymalizować takie parametry jak m.in. EP, CE, DP, CXP, Q1 mass, Q3 mass i pary MRM. W kolejnym rozdziale opisano proces wyboru kolumny do rozdziału chromatograficznego. Następnie zostały przedstawione zoptymalizowane parametry rozdziału chromatograficznego oraz podane inne niezbędne parametry potrzebne do odtworzenia opracowanej metody wraz z uzyskanymi wynikami. Dodatkowo opisano proces walidacji opracowanej metody.
The presented dissertation concerns the development of a quick and simple method of identifying new psychoactive substances using the LC-MS/MS technique with the use of SCIEX Exion LC liquid chromatograph with an autosampler of liquid samples coupled with a mass spectrometer with a triple quadrupole SCIEX Triple Quad 3500 analyzer.In the theoretical part, in the first place there is a description of new psychoactive substances, their classification, legal regulations, European and Polish statistics are also given, and the groups of tryptamines, synthetic cathinones and cannabinoids are described in more detail, because the representatives of such groups are most frequently studied in this study. Then, the focus was on methods used for the separation and qualitative analysis of compounds, i.e. liquid chromatography, mass spectrometry and the combination of these two techniques, i.e. LC-MS/MS. Finally, examples of applications of this method in forensic science and, in particular, the detection of NPS are described.The experimental part first describes the purpose of the research, the equipment used, the reagents used and the method of preparation of the test solutions. Then the process of optimizing the mass spectrometer was presented, during which such parameters as, among others, EP, CE, DP, CXP, Q1 mass, Q3 mass, and MRM pairs, were optimized. The next chapter describes the process of selection of a column for chromatographic separation. Then, the optimized parameters of the chromatographic separation were presented as well as other necessary parameters needed to reproduce the developed method along with the obtained results. Additionally, a validation process of the obtained method was described.
